Characterization of 40,000- and 25,000-dalton intermediate precursors to Rauscher murine leukemia virus gag gene products

Abstract: Under steady-state labeling conditions, Rauscher murine leukemia virus-infected NIH Swiss mouse cells contain at least three major polyproteins derived from the viral gag gene. They have molecular weights of 65,000, 40,000, and 25,000. They have been termed pPr65gag, Pr409a9, and pPr259'9. pPr659ag has been shown by a number of laboratories to be composed of all four core proteins (pl5, ppl2, p30, and plO). In this paper, Pr40ga was found to contain p30 and plO antigenic determinants and peptide sequences, whe… Show more

“…Referring to the domains defined in MoMLV, the conserved sequences in Figure 2 lie within the CA domain. The CA protein of MoMLV is ∼30 kDa, spanning 263 amino acids, and forms the hydrophobic internal core of the viral virion (32,33). The MoMLV sequence corresponding to amino acids 27-217 in the p30 protein (amino acids 243-433 in the MoMLV gag protein) is represented in Figure 2, demonstrating that similarity between MoMLV and chromoviruses can be recognized in most parts of the p30 protein.…”

“…It most likely represents a partially cleaved gag product. Such a 40-kDa gag intermediate containing p30 and plO determinants has already been identified in MuLV (33). This p4W'ag' intermediate most likely arises from the helper Pr659"-' precursor.…”

“…Anti-p30 (lane 3), anti-p 12 (lane 4), anti-p 15 (lane 5), and anti-p 10 (not shown) precipitate p36 in different amounts, which may reflect antibody titer or avidity differences. As observed by others (28), the heteroantisera reactive with p 10, p 12, and p 15 may be contaminated with low levels of antibody to p30, therefore, the precise antigenic determinants present on p36 are ambiguous. However, the observation that anti-pl2 (lane 4)precipitates larger amounts of p36 than of p30 suggests that its reactivity with p36 is not due solely to these contaminating anti-p30 antibodies.…”
