Characterization of a Novel Panton-Valentine Leukocidin (PVL)-Encoding Staphylococcal Phage and Its Naturally PVL-Lacking Variant

Abstract: A new siphophage (LH1) was isolated from raw milk using a Staphylococcus aureus ST352 host. Its genome (46,048 bp, 57 open reading frames) includes the two genes encoding Panton-Valentine leukocidin (PVL), a virulence factor usually harbored by S. aureus prophages. Nine structural proteins were identified, including a tail protein generated through a ؉1 frameshift. A phage lytic mutant was isolated, and its analysis revealed the deletion of genes coding for the PVL and an integrase. The deletion likely occurre… Show more

“…Phage genomes maintain a mosaic structure and gene exchange might occur by horizontal gene transfer (Dekel-Bird et al , 2013; El Haddad & Moineau, 2013; Labrie et al , 2013), which could then cause the diversity in the phage genome structure. The host lysis genes of the Wβ-group phages were quite different either in the amino acid sequence or in the type of the endolysin (Kikkawa et al , 2008; Yuan et al , 2012a).…”

“…A single copy of a DR that has been implicated in the deletion of the lukSF-PV and integrase module [43] was found downstream of the lukSF-PV genes in all prophages however, a second copy was not be found in any of the genomes. Nine lukSF-PV SNPs were found, six in lukS-PV and three in lukF-PV and there were eight SNP profiles ( Table 2).…”

“…Associations with osteomyelitis and pyomyositis are now strongly suggested [117][118][119][120][121][122][123]. The locus encoding PVL is inside related bacteriophages [124][125][126][127]128]. But the recent emergence of methicillin-resistant strains (MRSAs) producing this toxin may confer an increased morbid risk and necessitates careful therapeutic attitudes [129][130][131].…”
