Characterization of avian myeloblastosis-associated virus DNA intermediates

Abstract: The major species of unintegrated linear viral DNA identified in chicken embryonic fibroblasts infected with either the avian myeloblastosis-associated viruses (MAV-1, MAV-2) or the standard avian myeloblastosis virus complex (AMV-S) has a mass of 5.3 x 106 daltons. An additional minor DNA component observed only in AMV-S-infected cells has a mass of 4.9 x 106 daltons. The unintegrated linear viral DNAs and integrated proviruses of MAV-1 and MAV-2 have been analyzed by digestion with the restriction endonuclea… Show more

“…The positions of hybridization probes are shown underneath the provirus. The restriction map has been compiled from Bergmann et al (1980), Souza and Baluda (1980) and the unpublished data of D.Westaway. Therefore the diagram represents a consensus MAV provirus.…”

“…Therefore the diagram represents a consensus MAV provirus. The HindruI site 1.2 kb from the 3' end of the genome may not be present in all MAV isolates (Bergmann et al, 1980). The U5 probe has been described previously (Payne et al, 1981).…”

“…of EcoRI-cleaved DNA from uninfected or MAV-2-infected CEF. High-molecular-weight cellular DNA was digested with EcoRI, electrophoresed in 0.7%o agarose gels, blotted, and hybridized with 125I-labeled AMV-S RNA as described previously (4,15,16). Autoradiographs of Southern blots of KpnI-cleaved DNA from uninfected or MAV-2-infected CEF.…”

“…CEF cultures from a single 11-day-old C/E chicken embryo (Spafas, Inc.) negative for group-specific antigen and for chicken helper factor were infected with MAV-2 at a multiplicity of 1 to 5 infectious units per cell. High-molecular-weight (-3 x 107) cellular DNA from the MAV-2-infected CEF was extracted, precipitated by the method of Hirt, and purified as previously described (4,6,7,9).…”
