Characterization of cDNAs encoding isoforms of hamster 3 beta-hydroxysteroid dehydrogenase/delta 5-->4 isomerase

Abstract: Western blot analyses of various hamster tissues reveal high levels of expression of 3 -hydroxysteroid dehydrogenase (3 -HSD) in adrenal and liver, and moderate levels of expression in kidney. The expression in liver is sexually dimorphic; very high levels of protein are observed in adult male liver but very low levels are seen in the female liver. Three distinct cDNAs encoding isoforms of 3 -HSD were isolated from hamster cDNA libraries. The type 1 isoform is a highaffinity dehydrogenase/isomerase expressed i… Show more

“…This finding suggests that the non-endocrine tissues of Japanese eel potentially express the P450scc and P450c17 gene, however, the transcript abundance is low and below the detectable level of conventional RT-PCR as employed in this study. Multiple forms of 3β-HSD and its expression in a variety of tissues have been demonstrated in mammals, especially in rodents [35,36]. Therefore, the wide tissue-distribution of the 3β-HSD transcript does not appear to be unique in Japanese eel and 3β-HSD may be involved in reproductive physiology through the regulation of local and/or circulating steroid hormone levels in teleosts, as they are in mammals.…”

“…A study on the regulation of adrenal steroidogenic enzymes suggested that the hamster could be a good model for studying human steroidogenesis. With this in mind, three isoenzymes of 3␤-HSD were characterized in the hamster (96,97). The type I isoenzyme was isolated from an adrenal cDNA library and was identified as being a low K m 3␤-HSD (K m : PREG, 5.5 m; DHEA, 2.4 m).…”
