Osteogenic sarcoma (OGS) is a primary bone cancer, characterized by aggressive neoplasm from mesenchymal oncogenesis. However, the clinical therapeutic regimen against OGS is limited. Therefore, potential medication warrants to be further developed. Our previous study indicates that formononetin (FN) exerts effective pharmacological activity against OGS. This study aimed to further decipher the molecular mechanism behind this benefit. Patients with OGS were recruited for clinical data assay and immunoassay. Human OGS cell line (U2OS) and tumor‐bearing nude mice were subjected to a battery of biochemical analyses and immunoassays for integrative evaluation of FN‐exerted anti‐OGS effects. In human data, OGS samples showed increased expressions of ERα, p‐PI3KCATyr317, and p‐AKT Ser473 proteins, followed by notably upregulated miR‐375 content in comparison with that in OGS‐free. In addition, FN‐treated U2OS cells showed inhibited cell proliferation, elevated lactic dehydrogenase production and lowered endogenous miR‐375 level in cells. Further, reduced immunopositive cells of Ki‐67, p‐PI3KCA Tyr317, and p‐AKT Ser473 were observed by the treatments of FN, while the intracellular Bax‐ and Apaf‐1‐positive cells were increased dose‐dependently. Beneficially, FN‐treated tumor‐bearing mice exhibited reduced tumor mass and intercellular miR‐375 expression. Meanwhile, immuno‐labeled cells and proteins of Bax, Caspase‐3, and Apaf‐1 in FN‐treated mice were increased dose‐dependently, whereas ERα, p‐PI3KCA Tyr317, and p‐AKT Ser473 positive cells and proteins were downregulated, respectively. Collectively, our current results elucidate that FN exerts effective therapeutic benefits against OGS, and the pharmacological mechanism may be related to promoting cell apoptosis by inactivating intracellular miR‐375/ERα−PI3K/AKT cascaded pathway.

Anti‐colorectal cancer targets of resveratrol and biological molecular mechanism: Analyses of network pharmacology, human and experimental data

In this study, colorectal cancer (CRC)-diseased targets and resveratrol (Res)associated targets were combined and constructed by the use of grouped databases for identification of the predicted targets. After production of target-functional protein interaction network of Res anti-CRC, the topological analysis was used to create the core targets of Res anti-CRC. All core targets performed the analyses of biological function and pathway enrichment to optimize the biological processes and key signaling pathways of Res anti-CRC. The resultant five core therapeutic targets of Res anti-CRC were identified as protein kinase B1 (AKT1), interleukin 6 (IL6), Tumor protein p53 (TP53), vascular endothelial growth factor, and mitogenactivated protein kinase 1, respectively. Biological processes of Res anti-CRC were predominantly associated with regulating apoptosis, immune response, cellular communication, signal transduction, and metabolism of the nuclide. In addition, the top 10 key signaling pathways were identified, respectively. In human CRC sample assays, CRC histologic sections showed elevated expression of AKT1 and IL6 proteins, accompanied with abnormal changes in blood molecules. In pharmacological experiments of Res anti-CRC in vitro, Res-treated HCT116 cells showed inhibited cell growth, induced cell death. In addition, downregulation of intracellular AKT1 and IL6 expression were checked in Res-treated HCT116 cells.Taken together, these bioinformatic findings and preliminary validated data uncovered pharmacological molecular mechanisms associated with Res anti-CRC, and further identified top five core therapeutic targets. Hepatic function is of great concern in metabolic and immunological homeostasis.Traditionally, medical management to liver damage may benefit from phytomedicine, such as Chinese herbs. In southern China, Polygonum perfoliatum L can contribute to alleviating pathological symptoms of liver disease, such as hepatitis.However, bioactive compounds of hepatoprotection in this herb are still less to be investigated. In this study, clinical data of patients with drug-induced liver injury were collected on the basis of serological analyses. In addition, we extracted and identified total flavonoids from Polygonum perfoliatum L (TFPPL) before implementing biochemical experiments in vivo. In human data, the blood contents of liver function enzymes were visibly elevated, and the percentage of immune cells were abnormally changed. The data from the animal study showed that TFPPL-treated carbon tetrachloride-exposed mice resulted in reduced absolute liver mass and lowered blood levels of liver functional enzymes (alanine transaminase and aspartate transaminase). In enzyme-linked immunosorbent assay, the comparable data indicated that serological tumor necrosis factor α (TNF-α), interleukin 6, and heat shock protein 90 (Hsp90) contents were reduced in TFPPL-treated mice. In histopathological observations, TFPPL-treated mice exhibited reduced hepatocellular Hsp90, TNF-α, nuclear factor κ-light-chain-enhancer of activated B cells-p65 positive cells, and lowered Bax and caspase-3-labeled cells in the livers. Further, intrasplenic integrin β1, 5′-nucleotidase, and antigen KI-67 positive cells were increased after TFPPL treatments. Taken together, our present findings demonstrate that herbextracted TFPPL exert potential hepatoprotective activities against chemical-induced liver damage in mice, possibly through the pharmacological mechanisms of inhibiting inflammatory stress and apoptosis, inactivating Hsp90 bioactivity in the liver, and improving splenic immunocompetence.
