Abstract:The productivity of arid legumes, such as Clusterbean (Cyamopsis tetragonoloba), Cowpea (Vigna unguiculata), Moth bean (Vigna aconitifolia) and Horse gram (Macrotyloma uniflorum), may remain stagnant over decades because of their high susceptibility to root diseases. Besides, there is a limitation on the information about molecular diagnosis and intraspecific genetic variability of root pathogens in arid legumes. To contribute in this field, we assessed a total of 52 isolates from 88 root samples that were found infected with fungal pathogens in Jodhpur, Jaipur and Bikaner Districts of Rajasthan. Diseased roots samples were analyzed following standard microbiological methods for fungus extraction and purification, and for genetic studies. Irrespective of the geographical location from where the diseased samples were collected, all pathogen isolates were clustered in RAPD dendrograms as per their respective genera. Phylogram, based on multiple sequence alignment, revealed that different genera (i.e. Fusarium, Neocosmospora and Syncephalastrum), separated from each other, and species within the same genera, clustered together with their reference sequences with apreciable bootstrap values. Out of 20 representative isolates representing each cluster and all outgroups sequenced, eight were molecularly identified as Neocosmospora vasinfecta, five as Fusarium solani, two as Neocosmospora striata, two as Fusarium acutatum, one as Syncephalastrum monosporum, one as Fusarium oxysporum and one as Fusarium species. The root pathogens of the arid legumes were found neither restricted to a geographical location nor were host specific in nature. Fusarium solani wilt in cowpea and seedling rot in moth bean, F. oxysporum wilt in moth bean, F. acutatum damping off in cowpea and Clusterbean, Fusarium sp. seedling rot in Clusterbean, Neocosmospora striata root rot in cowpea and wilt in Clusterbean and Syncephalastrum monosporum root rot in Clusterbean were molecularly identified as new fungal records as pathogens causing root diseases in arid legumes. 3O r i g i n a l b e i t r a g Zusammenfassung Der erreger der Zwiebelbasalfäule, Fusarium oxysporum f. sp. cepae, ist aufgrund seiner hohen temperaturansprüche vorwiegend in wärmeren anbauregionen verbreitet. Mittlerweile tritt er auch in Deutschland auf, insbesondere in Jahren mit höheren Durchschnittstemperaturen während der Vegetationszeit. Phytopathologische Untersuchungen an 300 symptomlosen Zwiebelbulben zeigten, dass ca. 10 % der bulben mit Fusarium spp. infiziert waren, wobei die art F. oxysporum dominierte. Steckzwiebeln, die auf diesen Flächen als Pflanzgut eingesetzt wurden, waren zu 19 bis 98 % latent mit F. oxysporum befallen. Das kontaminierte Pflanzgut führte bis zur Ernte jedoch nicht wie erwartet zu einem entsprechend hohem Auftreten an Pflanzen mit für die Zwiebelbasalfäule charakteristischen Symptomen wie nass-oder trockenfäule. es ist davon auszugehen, dass die Symptomausprägung insbesondere von den gegebenen klimatischen bedingungen beeinflusst wird. Dies unterstützen Ergebnisse zu Untersuchungen der Pathogenität isolierter Fusarium spp.-isolate unter kontrollierten Kulturbedingungen. So resultierte eine inokulation des Kultursubstrates mit ausgewählten Fusarium spp.-isolaten unter für den Zwiebelanbau suboptimalen Kulturbedingungen in einer Reduktion des Pflanzenaufgangs um bis zu 70 %, während dies unter optimalen Kulturbedingungen nicht zu beobachten war. Unter optimalen bedingungen wurde jedoch in späteren entwicklungsstadien eine Reduktion des Pflanzenwachstums festgestellt. neben F. oxysporum wurde sowohl an den Zwiebeln und insbesondere am Saatgut F. proliferatum nachgewiesen. Dieser gleichfalls wärmeliebende Pilz kam in allen untersuchten Saatgutproben vor, wobei Kontaminationsraten bis zu 62 % ermittelt wurden. Für F. oxysporum wurde dagegen nur eine Befallshäufigkeit von unter 1,5 % an einigen Saatgutproben ermittelt. Fusarium basal plate rot caused by Fusarium oxysporum f. sp. cepae is an important disease of onion in many countries of the world. A total of 116 single spore isolates of Fusarium oxysporum f. sp. cepae representing seven populations from Turkey and one population from Colorado, USA were subjected to molecular marker analysis using Random Amplified Polymorphic DNA (RAPD) technique. Using eleven polymorphic primers, 110 RAPD fragments were obtained with an average of 10 polymorphic bands per primer. Cluster analysis with UPGMA revealed five distinct lineages at arbitrary level of 65% similarity. The majority of F. oxysporum f. sp. cepae isolates from Turkey were observed to derive probably from the same clonal lineage. Genetic estimates and population differences demonstrated that the isolates from Turkey were significantly distinct from Colorado isolates and that Bursa population was the most divergent among Turkish populations. Cluster analysis of Nei's genetic distances supported populations grouping according to the geographical regions. Comparison of genetic differentiation estimates (H T : 0.140, H S : 0.124) revealed low levels of genetic differentiation among Turkish populations. Only, 11.4% of total genetic diversity (G ST ) attributed to differentiation among the geographical populations. Analysis of molecular variance (AMOVA) confirmed that there was low genetic differentiation among populations. The results suggest that RAPD-PCR is a useful method for analyzing genetic variation within and between populations of F. oxysporum f. sp. cepae.
