Characterization of Fusarium oxysporum f.sp. cepae from Onion in Turkey Based on Vegetative Compatibility and rDNA RFLP Analysis

Abstract: Seventy-five isolates of Fusarium oxysporum f.sp. cepae, the causal agent of basal plate rot on onion, were obtained from seven provinces of Turkey. The isolates were characterized by vegetative compatibility grouping (VCGs) and restriction fragment length polymorphism (RFLP) analysis of the nuclear ribosomal DNA intergenic spacer region (IGS). Forty-eight vegetative compatibility groups were found, each containing a single isolate. Only one isolate formed strong heterokaryons with the reference isolates of VC… Show more

“…A broad pathogenic and genetic diversity was detected among isolates of Fusarium oxysporum f. sp. cepae restriction digestions of IGS regions of rDNA classified the isolates from Turkey and Colorado into 13 groups (Bayraktar, Türkkan, & Dolar, 2010;Mess et al, 1999) suggested that F. oxysporum f. sp. lycopersici is a polyphyletic taxon.…”

“…Also, the identification was confirmed by PCR using species-specific FOF/R 1 primers, producing a single fragment of 340-bp (Mishra et al 2003). The pathogenicity of isolates was determined by seedling and bulb inoculation methods used in a previous study (Bayraktar et al 2010). Pathogenic variability among the isolates was found to vary from 1 to 5, most of them were highly virulent on onion plants.…”
