Characterization of Oxytocin, Vasopressin, and Neurophysin from the Bovine Corpus Luteum*

Abstract: Acid extracts of corpora lutea collected from nonpregnant cows were found to contain oxytocin, arginine vasopressin, and neurophysin. The inhibition curves of the oxytocin and vasopressin extracts showed parallelism with the appropriate standard preparations in specific RIAs and eluted at the same position as the standards using high performance liquid chromatography (HPLC). The neurophysin extract showed parallelism in a bovine neurophysin I RIA and had a similar elution position to the standard on both Sepha… Show more

“…Measurements were made on the same extracts used for the oxytocin assay, using the specific radioimmunoassay described previously by Wathes et al (1983), using an antiserum (84/1) raised and charac¬ terized in the Department of Anatomy. This antiserum cross-reacts <6% with lysine vasopressin and arginine vaso¬ tocin, and <003% with oxytocin, mesotocin, and the neurophysins.…”

“…These results suggest that oxytocin may be derived from the corpus luteum (CL) and be involved in the process ofluteolysis. Oxytocin has been detected in the CL of sheep , cows (Wathes et al, 1983) sheep and women by use of high performance liquid chromatography, milk ejection and rat uterus strip bioassays and radioimmunoassays (RIA) . However the RIA employed are open to criticism on the grounds that immunological activity can be separated from biological activity, and the RIA may measure degraded and biologically inactive fragments of OT (Kumaresan et al, 1969).…”
