Nociceptive Transient Receptor Potential channels such as TRPV1 are targets for treating pain. Both antagonism and agonism of TRP channels can promote analgesia, through inactivation and chronic desensitization. Since plant-derived mixtures of cannabinoids and the Cannabis component myrcene have been suggested as pain therapeutics, we screened terpenes found in Cannabis for activity at TRPV1. We used inducible expression of TRPV1 to examine TRPV1-dependency of terpene-induced calcium flux responses. Terpenes contribute differentially to calcium fluxes via TRPV1 induced by Cannabis-mimetic cannabinoid/terpenoid mixtures. Myrcene dominates the TRPV1-mediated calcium responses seen with terpenoid mixtures. Myrcene-induced calcium influx is inhibited by the TRPV1 inhibitor capsazepine and Myrcene elicits TRPV1 currents in the wholecell patch-clamp configuration. TRPV1 currents are highly sensitive to internal calcium. When Myrcene currents are evoked, they are distinct from capsaicin responses on the basis of I max and their lack of shift to a pore-dilated state. Myrcene pre-application and residency at TRPV1 appears to negatively impact subsequent responses to TRPV1 ligands such as Cannabidiol, indicating allosteric modulation and possible competition by Myrcene. Molecular docking studies suggest a non-covalent interaction site for Myrcene in TRPV1 and identifies key residues that form partially overlapping Myrcene and Cannabidiol binding sites. We identify several non-Cannabis plantderived sources of Myrcene and other compounds targeting nociceptive TRPs using a data mining approach focused on analgesics suggested by non-Western Traditional Medical Systems. These data establish TRPV1 as a target of Myrcene and suggest the therapeutic potential of analgesic formulations containing Myrcene.

Aims: Considering the lack of scientific studies focused on the pharmacological activity of Cordia millenii essential oil, this work was designed to evaluate the anti-inflammatory and anti-nociceptive activities of essential oil from the root bark. Study Design: The design of the study include collection of root bark of Cordia millenii, hydrodistillation of essential oil from the plant and evaluation of its anti-inflammatory and anti-nociceptive potentials. Place and Duration of Study: Department of Chemistry, Lagos State University, Nigeria between May 2017 and April 2018. Methodology: The root bark of C. millenii were collected from Ayetoro, Ilesha (7°37¢0N 4°43¢0E), Osun State, Nigeria in June 2017. Essential oils were obtained from the air-dry sample by hydrodistillation procedure in an all glass Clevenger-apparatus. The anti-inflammatory activity was evaluated by carrageenan-induced rat paw edema. The anti-nociceptive action was established from the hot-plate analysis. Statistical analysis was performed using GraphPad Prism (version 7.02). Results: The anti-inflammatory activity of the essential oil was statistically significant (p < 0.001) at 1st and 3rd h for the 100 mg/kg p.o., at 1st-3rd h for the 200/kg mg p.o. and at 3rd and 4th h for the 400 mg/kg p.o. In addition, the 100 mg/kg p.o. showed significant activity (p < 0.01) at 2nd h. Also, the anti-inflammatory activity was significant (p < 0.05) for 100 mg/kg p.o. (4th h), 200 mg/kg p.o. (4th h) and 400 mg/kg p.o (1st h). The essential oil of C. millenii displayed high activity (p <0.001) for all doses in the hot plate anti-nociceptive assay which was time and dose independent. Conclusion: Results demonstrate that the essential oil of C. millenii was effective in the treatment of inflammatory conditions, thereby supporting the traditional use of this herb.

For centuries, essential oils (EOs) have held a significant place in traditional medicinal practices worldwide and are commonly known for their aroma and taste. Among the 3000 recognized types, only 300 have commercial use in the food and pharmaceutical sectors. These potent natural extracts, derived from aromatic plants and herbs, have garnered attention not only in ancient healing traditions but also within modern health‐care systems. Their rich history is marked by a diverse array of therapeutic properties that have made them indispensable tools for promoting health and well‐being. Additionally, the utilization of EOs as food preservatives is an innovative and natural approach in extending the shelf life of various food products. In addition, EOs are beneficial in the treatment of cancer and other diseases. Because of their outstanding biological activities and health benefits, research on EOs has exploded in recent years. In present work, an intensive review was done on published data on therapeutic potential of some major EOs and their utilization as a preservative in food industries. Meanwhile, an investigation of the methods specifically used for the separation of EOs and regulatory considerations are also reported. The article concludes with comments on future trends.
