The molecular structures of the radical scavenging compounds present in extracts of Potentilla fruticosa blossoms were elucidated and the antioxidant activities of various extracts were determined. The activities of the different fractions were monitored by off-line and on-line RP-HPLC DPPH • and ABTS •+ scavenging methods. Twelve compounds were isolated and identified, namely ellagic acid, catechin, quercetin-3-β-glucopyranoside, quercetin-3-β-galactopyranoside, quercetin-3-β-rutinoside, quercetin-3-β-glucuronopyranoside, quercetin-3-α-arabinofuranoside, kaempferol-3-β-rutinoside, kaempferol-3-O-β-(6 -O-(E)-p-coumaroyl)glucopyranoside, rhamnetin-3-β-glucopyranoside and rhamnetin-3-β-galactopyranoside. The radical scavenging activity of each isolated compound was measured using DPPH • and ABTS • + assays and compared with the activity of rosmarinic acid. Catechin and ellagic acid were found to be the most active radical scavengers. The antioxidant properties of plant fractions were assessed in model systems by measuring superoxide anion and hydrogen peroxide scavenging, β-carotene bleaching, hexanal production in edible oil, peroxide formation, and the increase in UV absorbance in the course of oxidation.

This report compared the phenolic compounds and antioxidant activity of the leaves, flowers, and stems of Potentilla fruticosa L. collected from two main production areas of P. R. China (Taibai Mountains and the Qinghai Huzhu Northern Mountains). The results indicated that there were significant differences in the phenol contents and antioxidant activities among the different organs and between the two productions. High-performance liquid-chromatography analysis indicated that hyperoside, (+)-catechin, ellagic acid, and rutin were the primary compounds in leaves and flowers; for stems, the content of six phenolic compounds, from two productions, were the lowest. The 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) di-ammonium salt (ABTS), ferric reducing power (FRAP), lipid peroxidation assays, and microbial test system (MTS) were used to evaluate the antioxidant activity. The results demonstrated that the leaves from two productions exhibited powerful antioxidant activity than other organs, which did not significantly differ from that of the positive control (rutin), followed by the flowers and stems. The correlation between the content of phytochemicals and the antioxidant activities of different organs showed that the total phenol, tannin, hyperoside, and (+)-catechin contents may influence the antioxidant activity, and these compounds can be used as markers for the quality control of P. fruticosa.

Total of 35 phenolics and flavonols were isolated from flowers and leaves of Dasiphora fruticosa Rydb., from branches of Myricaria alopecuroides Schrenk. and from the herb of Sedum hybridum L. The isolated compounds were identified on the basis of spectral data and tested for their anti-oxidative, acetylcholinesterase and pancreatic lipase inhibitory activities.Quercetin glycosides, gallic acid, (-)EGCG and gossypetin-8-O-xylopyranoside tellimagrandin II exhibited strong anti-oxidative activity by the DPPH scavenging method. The acetylcholinesterase inhibitory activity of quercetin glycosides and (-)EGCG was higher than those of other compounds. Whereas, tellimagrandin II, (-)EGCG and gallic acid derivatives exhibited the most potent inhibitory activity against the pancreatic lipase enzyme among the isolated compounds. Only (-)EGCG showed a prominent activity against all assayed experiments. It was concluded that these plants could be studied further for their potential as anti-oxidative, anti-aging and lipid lowering active products.
