Chemical mutagenesis testing in Drosophila. III. Results of 48 coded compounds tested for the national toxicology program

Abstract: Forty-eight chemicals were tested for mutagenicity in Drosophila melanogaster. Each compound was tested first for the induction of sex-linked recessive lethals (SLRLs) by feeding the chemical in a solution of 5% aqueous sucrose. If it was not mutagenic by this route, the chemical was tested by injection in a solution of 0.7% aqueous NaCl. If found inactive, the compound was not tested further. If it was mutagenic, the compound was tested for the induction of reciprocal translocations (RTs) using the exposure p… Show more

“…The assay with CHO cells did meet FIFRA guidelines; however, it was unclear if the sister chromatid exchange assay met FIFRA guidelines due to insufficient information. One sex-linked recessive lethal (SLRL) assay in Drosophila melanogaster had equivocal results (Valencia et al, 1985), but there was also insufficient information to determine if this study met FIFRA guidelines.…”

“…A sex-linked recessive lethal assay was conducted in which Drosophila melanogaster Canton-S wild-type males were fed chloropicrin (91% pure) at 0 and 150 ppm for 4 hours or injected at 0 and 100 ppm (Valencia et al, 1985). Males were then mated with 3 harems of Basc virgin females to produce 3 broods of 3, 2, and 2, days.…”

“…Our procedures for the SLRL assay have been described elsewhere [Woodruff et al, 1984Valencia et al, 1985;Yoon et al, 1985;Zimmering et al, 19851. Briefly, each chemical was tested for the ability to induce recessive lethals after being fed to adult males. If no significant mutation induction was observed, the compound was retested after injection.…”

“…HCTZ, administered either by injection or in the feed, did not induce sex-linked recessive lethal mutations in Drosophila melanogaster [Valencia et al, 1985]. HCTZ was also negative for micronucleus and sister chromatid exchange induction in Chinese hamsters, negative in the mouse spermatogonia assay, and negative in the mouse dominant lethal assay [EMEA, 1999].…”
