Chemokine-Induced Macrophage Polarization in Inflammatory Conditions

Abstract: Macrophages represent a heterogeneous cell population and are known to display a remarkable plasticity. In response to distinct micro-environmental stimuli, e.g., tumor stroma vs. infected tissue, they polarize into different cell subtypes. Originally, two subpopulations were defined: classically activated macrophages or M1, and alternatively activated macrophages or M2. Nowadays, the M1/M2 classification is considered as an oversimplified approach that does not adequately cover the total spectrum of macrophag… Show more

“…However, this classification is useful in providing an overview of the diverging cellular responses to CCL2. Multiple cytokines have been described in the complex network of macrophage polarization, and their roles have been successively revealed (151,152). The M1 phenotype is characterized by increased expression of cytokines, such as TNFα, IL-1β, IL-6, and IL-12; increased expression of surface receptors, such as Fcy-RI, II, and IIIA; as well as the production of reactive oxygen species (ROS) and reactive nitrogen species (151,153).…”

“…Excessive production of proinflammatory mediators from these macrophages might have effects on the parasites, but could also lead to excessive injury of surrounding tissue and hence result in extensive periductal fibrosis. As alternatively activated (M2) macrophages are known to play an important role in wound healing and acquire a pro-fibrotic phenotype (27), our MDM activities reported here may be predominantly generated by M2 macrophages. More in-depth mechanisms on macrophage sub-type responses are required.…”

“…Actin cytoskeleton and cell adhere, are closely related to epithelial to mesenchymal transition (EMT) and cell migration (39,40). Chemokine can recruit macrophage and induced macrophage M2 polarization which promoted tumor lymphatic metastasis (41). MAPK and TGF-beta signaling pathway has been known to mediate cell EMT and migration in thyroid carcinomas (42,43).…”
