Chemopreventive effects of vanadium toward 1,2‐dimethylhydrazine‐induced genotoxicity and preneoplastic lesions in rat colon

Abstract: In the present study, we have evaluated the antitumor effects of vanadium by monitoring DNA damage and chromosomal aberrations (CAs) during the early preneoplastic stage of 1,2-dimethylhydrazine (1,2-DMH)-induced colon cancer in male rats. Treatment with 20 mg/kg 1,2-DMH for 6 weeks resulted in the formation of aberrant crypt foci (ACF), a putative preneoplastic lesion associated with colon cancer development, while cotreatment with ammonium monovanadate (0.5 ppm in the drinking water) reduced ACF formation by… Show more

“…Recent data indicate spermicidal and anti-HIV activity of vanadium compounds (D'Cruz et al 2003) and the use of these compounds for the treatment of diabetes (Heinemann et al 2003;Sakurai et al 2004;Yang et al 2004) as well as the prevention of animal carcinogens (Kanna et al 2004;Molinuevo et al 2004). It was reported that decavanadate (V 10 ) is the major protein-bound species of vanadium and have a stronger effect on various enzymes, when compared to other vanadate oligomers (Aureliano and Gândara 2005).…”

“…It has been reported that vanadate reverses drug resistance [6,7], increases glucose metabolism [8] and influences the activity of several enzymes, either inhibiting or stimulating [9][10][11]. Recent data associated vanadium essentially to the treatment of diabetes, due to its insulin mimetic properties [12][13][14] and to the prevention of animal carcinogens [15,16], however some studies of DÕCruz and its collaborators have reported spermicidal and anti-HIV activity of vanadium compounds [17].…”

“…Moreover, vanadium toxicological effects in several cell types have been related with ROS production [16][17][18][19]. Nevertheless, vanadium was recently reviewed as ''an element of atypical biological significance'' [20], due to insulin mimetic properties of vanadium [16,21,22] and to the preventive effects of vanadium against animal carcinogens [23][24][25][26].…”
