Chicken egg yolk immunoglobulin (IgY) developed against fusion protein LTB-STa-STb neutralizes the toxicity ofEscherichia coliheat-stable enterotoxins

Abstract: Aim: To obtain a recombinant enterotoxigenic Escherichia coli (ETEC) fusion enterotoxin protein LTB-STa-STb (Bab) that can express the immunogenicity of the haptens STa and STb and induce their corresponding neutralizing antibodies. Methods and Results: The three important ETEC enterotoxin genes coding LTB, STa and STb were PCR-amplified, and the amplified products were fused to construct the trivalent enterotoxin expression vector pET30-Bab. SDS-PAGE and Western blot were used to verify the expression of the … Show more

“…were able to neutralize the toxic effect of ST. In a suckling mouse model, anti-Bab IgYs were able to neutralize STa (2× dilution of IgYs) and STb (32× dilution of IgYs) ( 179 ). Another protein important for ETEC invasion is K99 fimbrial protein that enables colonization of the host epithelium.…”

“…The polyvalent DNA vaccine pCI-STa-LTB-STb expressed the STa-LTB-STb fusion peptide in vitro in cultured Hela cells. These egg yolk antibodies were able to neutralize the natural toxicity of STa and LTb with the highest dilution of 1/2 and 1/32 in a suckling mouse assay [ 45 ]. These results indicate that the recombinant STa-LTB-STb protein has the potential to serve as an effective and convenient polyvalent toxoid which can provide broad protection against ETEC-induced diarrhea.…”
