Cholesterol Contributes to Male Sex Differentiation Through Its Developmental Role in Androgen Synthesis and Hedgehog Signaling

Abstract: Two specialized functions of cholesterol during fetal development include serving as a precursor to androgen synthesis and supporting hedgehog (HH) signaling activity. Androgens are produced by the testes to facilitate masculinization of the fetus. Recent evidence shows that intricate interactions between the HH and androgen signaling pathways are required for optimal male sex differentiation and defects of either can cause birth anomalies indicative of 46,XY male variations of sex development (VSD). Further, … Show more

“…Our data are consistent with studies performed in experimental rodent and human liver disease (57)(58)(59), showing that injuryinduced release of Hedgehog ligands from epithelia leads to hepatic myofibroblast activation. Studies in other organs in humans, mice, zebrafish, and fruit flies also support the ability of Hedgehog signaling to drive local and remote tissue development, cancer, and post-injury fibrosis (60)(61)(62)(63)(64)(65). The literature on cardiac Hedgehog signaling has focused mainly on the beneficial impact of Hedgehog signaling in cardiac development and on successful scar formation in the first 7 days after myocardial infarction (66).…”

“…Epidemiological and preclinical observations have suggested that cholesterol-lowering plays a role in reducing the development and progression prostate cancer ( Alfaqih et al, 2017 ; Revilla et al, 2021 ). Cholesterol is an important precursor for de novo androgen synthesis ( Kothandapani et al, 2021 ). Similarly, androgen also plays a critical role in the pathogenesis and development of prostate cancer ( Dai et al, 2017 ).…”

“…Androgens are first produced by the Leydig cells in the fetal testis, in which express the key steroidogenic enzymes for androgen production Cytochrome P450 17a1 ( Cyp17a1 ) and 3β-Hydroxysteroid (3β-HSD) ( 11 ). These two enzymes and other enzymes in the cascade of steroidogenesis convert cholesterol to androgens ( 11 , 12 ). The testis-derived androgens then enter the blood circulation and reach the target organs.…”
