Diabetes mellitus (DM) causes an imbalance in the oxidative status of the human body. Three novel Dapagliflozin (Dapg) Zn (II), Cr (III) and Se (IV) complexes were prepared and characterized by elemental analysis, IR, electronic spectra, magnetic susceptibility, scanning electron microscopy (SEM) and X-ray diffraction. The molar conductance values confirmed the non-electrolytic nature of the Dapg complexes. According to spectral data, Dapg acts as a bidentate ligand. The thermal analyses of the complexes were studied using the DSC technique. The surface morphology and particle sizes of the Dapg complexes were investigated using SEM and XRD. XRD confirmed the crystalline structure for the complexity. This study investigated the effect of novel metal complexes of Dapg with the metals Zn (II), Cr (III) and Se (IV) on oxidative injury and tissue damage in the hepatic tissue of streptozotocin (STZ)-induced diabetic male rats. DM was experimentally induced in male rats. The diabetic rats received Dapg, Dapg/Zn, Dapg/Cr and Dapg/Se orally for 30 successive days. Male rats exposed to STZ showed multi-histopathological alterations in their hepatic tissue, including inflammatory and structural changes. STZ elevated oxidative stress markers in the hepatic tissue and lowered the antioxidant defense enzymes. Supplementation of Dapg with Zn, Cr or Se novel complexes significantly prevented hepatic injury and suppressed the generation of reactive oxygen species. The Dapg/Zn complex was highly effective against Bacillus subtilis and Streptococcus penumonia, while Dapg/Cr was highly effective against Escherichia coli and Pseudomonas aeruginosa, and Dapg/Se was highly effective against Staphylococcus aureas. In conclusion, Dapg novel metal complexes with Zn, Cr or Se protect against oxidative injury and the pathophysiological and bacterial complications of DM and hepatic tissue injury. The Dapg novel metal complexes improved hepatic functions, reduced blood glucose levels and enhanced the levels of antioxidant defense enzymes in diabetic male rats.

Background: Diabetes is a long-term illness characterized by high blood sugar levels. It is estimated that by 2045, there will be nearly 693 million diabetic patients worldwide, with half of the population remaining undiagnosed. Metformin, insulin, sulfonylureas, and thiazolidinediones were related to several risk factors, including hypoglycemia, bone fracture, weight gain, cardiovascular, renal, and other complications. In the present study, we have explored the DPP-IV inhibitors as a new class of antidiabetic drugs. Objectives: The goal of DPP-IV inhibitors is to raise levels of incretins (GLP-1 and GIP), which block glucagon release while boosting insulin secretion, slowing stomach emptying, and reducing blood glucose levels. Methods: A series of derivatives substituted on Oxadiazole of sulfonamide pyrrolidine were produced by reacting 1,2,4-oxadiazol and sulfonyl chloride at room temperature in the presence of ethanol and stirring until the reaction was complete. The compounds were characterized using IR, 1 H NMR, C 13 NMR, mass spectroscopy, elemental analysis, and screened for in vitro assay of DPP-IV inhibition. Results and Discussion: The IC 50 was calculated for compounds B-I, B-V, B-VI, B-XI, and B-XIII that inhibited enzymes significantly, IC 50 values ranging from 19.65 ± 2.60 nM to 11.32 ± 1.59 nM, respectively, and Vildagliptin (4.79±1.66 IC 50 nM) was used as a standard. The most active derivative substituted Oxadiazole of pyrrolidine sulfonamide is B-XI (11.32 ± 1.59 IC 50 nM) among all synthesized compounds. Conclusion: B-XI derivative has shown appreciable DPP-IVinhibitory action. The 1,2,4-oxadiazol-3-yl pyrrolidine-1-sulfonamide derivatives have shown anti-diabetic properties.

Ceftibuten dihydrate is a semisynthetic, orally administered, third generation cephalosporin antibiotic which is effective against most of the pathogens causing infections in the respiratory tract. Complexation of ceftibuten dehydrate (Ligand, L) was performed with hydrated Ni(II) salt (Metal, M) in the ratio of 2:1 (L:M) in aqueous medium at 90 oC. The metal complex was then characterized by spectral techniques and thermal analyses. The FT-IR spectral data of metal complex suggested the monodentate bonding of metal ion to carboxylate group. Spectral evidence also supported the formation of five-membered ring via coordination of metal ion to β-lactam nitrogen and carboxylate group of parent drug. Thermal behavior of ligand and complex were studied. Thus, thermoanalytical (DSC and TGA) results also supported the formation of new metal complex, indicating the successful interaction of metal ion to ligand. Dhaka Univ. J. Pharm.
