Chromosomal heterochromatin differentiation in Salmo trutta with restriction enzymes

Abstract: Fixed methaphase chromosomes of Salmo trutta were treated with different restriction enzymes. Each enzyme produces a specific banding pattern which demonstrates the value of restriction enzymes for chromosome banding iii this species. Digestion with Alul, DdeI, HaeIII, Hinfi and MboI indicated the existence of different classes of highly-repetitive DNA. The restriction endonuclease analysis carried out in Salmo trutta trutta and Salmo truttafario morphae has revealed no differences in respect of heterochromati… Show more

“…Current results also show that NORs in the European huchen are built with heterochromatin enriched in the restriction sites recognized by Dde I and Alu I. Similar observations have been reported in other fish species (SANCHEZ et al 1991;SALVADORI et al 1997;JANKUN et al 1998;DI et al 2006). Additionally, similar interindividual size heteromorphism of the NOR-bearing chromosome short arms observed in the European huchen was also recognized in the Siberian taimen (FROLOV & FROLOVA 2000).…”

“…2d). Some studies have been made to analyze the action of restriction enzymes on chromosomic preparations of fish, such as SANCHEZ et al, (1990SANCHEZ et al, ( , 1991, ABUIN et al (1994), BOUZA et al (1994), in which the use of the restriction enzyme Alu I also produced a linear differentiation similar to C banding, as observed in the species of fish analyzed here.…”

“…Great difficulties have arisen in the C-and G-banding studies of amphibians, and only in the last few years data on the sensitivity of chromo somes to the various banding techniques have been reported in teleosteans (Lloyd and Thorgaard, 1988;Cau et al, 1988Cau et al, .1992 Lozano et al, 1991;Sanchez et al, 1991; Hartley, 1991a. b; Padilla et al, I993;Vinaset al, 1994).…”

“…Data on tele osteans have accumulated, contributing to the elucidation of the role of heterochromatin components in evolutionary phe nomena (Gold et al. 1986;Sanchez et al, 1991). In view of this, we undertook a study of the effects of several RE, as well as of heterochromatin and NOR localization, in two cogeneric species belonging to the superorder Batoidea in order to get information on the localization of particular DNA sequences with different functional roles.…”
