Chronic stress causes neuroendocrine-immune disturbances without affecting renal vitamin D metabolism in rats

Abstract: Our data suggest that the stress-induced neuroendocrine-immune disturbances may account for the development of depression, but are not responsible for the insufficient VD status that frequently observed in depressed patients.

“…The observed noncorresponding changes in the activity, protein, and mRNA levels of the investigated CYP isoforms suggest some posttranscriptional or posttranslational modifications in CYP expression produced by CMS in the liver. The observed changes in CYP expression and activity may be caused by the ability of CMS to produce alterations in the function of the brain nervous system (Willner, 2016), which leads to the release of glucocorticoids and catecholamines, and modifications in the immune system (Kvetnansky et al, 2009;Jiang et al, 2014;Konstandi et al, Fig. 4.…”

“…However, these effects may be modified by catecholamines (adrenaline, noradrenaline) acting positively via hepatocyte a 1 and b 1/2 adrenergic receptor through the cAMP response element-binding protein pathway, but negatively via pancreatic b 2 receptor through the insulin pathway (Konstandi, 2013) on CYP expression (Konstandi, 2013). Moreover, CMS-induced immune disturbances that lead to elevation of plasma proinflammatory cytokines (interleukin-6, tumor necrosis factor-a, and tumor necrosis factor-g) may additionally diminish enzyme expression (Zidek et al, 2009;Jiang et al, 2014).…”

“…Hippocampal neurons in individuals with 22q11.2 deletion may already be vulnerable before chronic stress due to several factors including vitamin D insufficiency. Vitamin D insufficiency combined with exposure to chronic stress may be sufficient to further reduce expression of VDR and CYP27B1/CYP27A1 in the hippocampus, leading to a decrease in local production of vitamin D. As suggested by Jiang et al, 78 over time reduced expression of VDR/CYP27B1/ CYP27A1 may leave hippocampal neurons vulnerable to GC-induced cell death and further increase the risk for pathologies such as anxiety, depression and the development of schizophrenia.…”

“…4 A recent series of experiments examining GC exposure in an animal model of depression lends support to this hypothesis, and suggests that hippocampal vitamin D signaling may be involved in a compensatory adaptive response to allostatic load induced by chronic stress. 32,77,78 Lower vitamin D levels associated with impaired cognitive function has been reported in a number of studies. 79,80,81 Similarly, for children in the current study, lower full-scale IQ and poorer perceptual reasoning was associated with lower vitamin D blood concentrations.…”

“…INE system is stereonetwork regulation system with high precision and high complexity through cytokine, hormone, and other chemistry substances [28,29]. Physical and chemical biological characteristic of medium and hormone secreted by neuroendocrine system and lymph gene, monocyte secreted by immune system, and existing receptor in INE system cell surface are gradually discovered with the molecule-biology development.…”
