Circular RNAs (circRNAs) have been proved have a major impact on tumorigenesis. Whereas, the specific regulation of circRNAs during lung cancer progression is still need further investigation. Here, we found hsa_circ_0078767 was lowly expressed in lung cancer. Through CCK-8, colony formation, EdU staining, and flow cytometer apoptosis assays. We found that up-regulation of hsa_circ_0078767 repressed lung cancer cells proliferation and facilitated cell apoptosis in vitro and in vivo. Furthermore, we identified the regulatory function of hsa_circ_0078767 on ferroptosis during the malignant development of lung cancer. Elevated iron, Fe2+, and ROS levels was found in hsa_circ_0078767 overexpressed lung cancer cells. Ferroptosis activator erastin reduces the cell viability, and the hsa_circ_0078767 overexpression reinforced erastin-induced inhibitory effect on lung cancer cells. Mechanically, hsa_circ_0078767 sponged miR-330-3p to affect p53 expression. Up-regulation of miR-330-3p and knockdown of p53 reversed the role of hsa_circ_0078767 overexpression on lung cancer cells. Taken together, hsa_circ_0078767 may be a new research highlights for inhibiting lung cancer by promoting ferroptosis in lung cancer cells.

Background: Osteosarcoma is the most common primary malignant bone tumor, mainly affecting children, young adults, and the elderly. It is an aggressive cancer with a poor prognosis, exhibiting low survival rates even with standard treatment. Recently, circular RNA molecules capable of influencing gene expression through various functions, with their main role being acting as microRNA sponges and reducing their intracellular expression, have been identified. Recent studies have linked circular RNAs to osteosarcoma development and progression. Therefore, the present study aimed to investigate the alteration in circular RNA expression during osteosarcoma development and progression. Methods: An integrative literature review was conducted from September 10 th to November 12 th , 2021, using the following databases: PubMed/MEDLINE, SCOPUS, Web of Science, OVID, and EMBASE. 129 full articles were included in the review. The obtained data were organized using a standardized data collection instrument, which included the following information: altered expression profile of circular RNAs, associated cancer hallmarks, clinical-pathological relationships of circular RNAs, and perspectives on the studied circular RNAs. Results: A total of 94 distinct circular RNAs were identified, predominantly showing an increased expression pattern. Approximately 91% of the studies that aimed to identify the mechanisms of action of circular RNAs highlighted the function of circular RNAs as microRNA sponges. The most associated cancer hallmarks with the identified circular RNAs were proliferative signaling induction, invasion and metastasis, and resistance to cell death. The altered expression of these circular RNAs generally correlated with a worse prognosis for patients, as evidenced by clinical features such as shorter survival, advanced Enneking and/or TNM stage, higher incidence of metastasis, larger tumor size, and increased chemoresistance. Conslusion: These findings indicate the significance of circular RNA molecules in osteosarcoma carcinogenesis, suggesting their potential as new prognostic and/or diagnostic biomarkers, as well as alternative therapeutic targets in the fight against osteosarcoma.

Increasing research has shown that the abnormal expression of circRNAs is closely related to tumorigenesis, apoptosis, and patient prognosis in cervical cancer. This study aimed to reveal the procancer role of circIL21R in cervical cancer and investigate its related molecular mechanisms. Bioinformatics analysis revealed that circIL21R promotes the progression of cervical cancer via the miR-1205/PTBP1 axis. CircIL21R expression was significantly greater in tumor tissue than in adjacent normal tissue, and higher circIL21R expression indicated shorter survival. We applied MTS assays, EdU assays, and Transwell assays to show that the overexpression of circIL21R promoted cervical cancer cell proliferation and invasion. Mechanistically, circIL21R promoted the expression of PTBP1 by sponging miR-1205. Moreover, rescue assays confirmed that regulating the expression of miR-1205 or PTBP1 could reverse the tumorigenic effect caused by circIL21R overexpression. In addition, circIL21R promoted the tumorigenesis of cervical cancer in vivo. In summary, our study demonstrated that circIL21R was highly expressed in cervical cancer and upregulated PTBP1 expression by acting as a ceRNA for miR-1205, making outstanding contributions to several malignant biological processes in cervical cancers, such as growth, proliferation, and invasion. CircIL21R is a potential biomarker for the diagnosis and treatment of cervical cancer.
