Objectives MicroRNAs are abundant in eukaryotic cells and play key roles in cancers. Circular RNAs (CircRNAs) served as the competing endogenous RNAs (ceRNAs) in mediating multiple cell processes. This study aims to define the role of CircRNA CircZNF609/miR‐134‐5p in glioma as well as the underlying regulating mechanism. Methods Relative expression of miR‐134‐5p, CircZNF609 and BTG‐2 mRNA was determined by quantitative real‐time PCR. Cell proliferation was analysed by CCK‐8 assay. Cell migration was assessed by cell wound scratch assay. The direct regulatory of miR‐134‐5p on BTG‐2 and CircZNF609 was verified by luciferase report gene assay. Key findings MiR‐134‐5p was significantly upregulated in glioma cells. The overexpression of miR‐134‐5p inhibited cell proliferation and migration of glioma cell U251 and U87. Reversely, knock‐down of miR‐134‐5p enhanced cell proliferation and migration. Both BTG‐2 and CircZNF609 are the direct targets of miR‐134‐5p, and their expression could be negatively regulated by miR‐134‐5p. CircZNF609 was significantly upregulated in U251 and U87 cells and acted as an oncogene to promote cell proliferation and cell migration of glioma cell U251 and U87. Conclusions These data proved that CircZNF609 served as a competing RNA to bind miR‐134‐5p that promoted BTG‐2 expression leading to reduced proliferation and migration of glioma cell.

Background/Aims: Prostate cancer (PCa) is one of the main cancers that damage males’ health severely with high morbidity and mortality, but there is still no ideal molecular marker for the diagnosis and prognosis of prostate cancer. Methods: To determine whether the differentially expressed circRNAs in prostate cancer can serve as novel biomarkers for prostate cancer diagnosis, we screened differentially expressed circRNAs using SBC-ceRNA array in 4 pairs of prostate tumor and paracancerous tissues. A circRNA-miRNA-mRNA regulatory network for the differential circRNAs and their host genes was constructed by Cytoscape3.5.1 software. Quantitative real-time polymerase chain reaction analysis (qRT-PCR) was performed to confirm the microarray data. Results: We found 1021 differentially expressed circRNAs in PCa tumor using SBC-ceRNA array and confirmed the expression of circ_0057558, circ_0062019 and SLC19A1 in PCa cell lines and tumor tissues through qRT-PCR analysis. We demonstrated that combination of PSA level and two differentially expressed circRNAs showed significantly increased AUC, sensitivity and specificity (0.938, 84.5% and 90.9%, respectively) than PSA alone (AUC of serum PSA was 0.854). Moreover, circ_0057558 was correlated positively with total cholesterol. The functional network of circRNA-miRNA-mRNA analysis showed that circ_0057558 and circ_0034467 regulated miR-6884, and circ_0062019 and circ_0060325 regulated miR-5008. Conclusion: Our results demonstrated that differentially expressed circRNAs (circ_0062019 and circ_0057558) and host gene SLC19A1 of circ_0062019 could be used as potential novel biomarkers for prostate cancer.

Background Pressure ulcer (PU) is a common complication of acute and long‐time hospitalization, especially in elderly patients. The incidence and prevalence of PU are swiftly fortifying. Currently, our purpose was to investigate the functional impacts of circ‐Ttc3 on PU. Methods HaCaT cells were pretreated under hypoxia condition. Cell counting kit 8 assay and flow cytometry were utilized to test HaCaT cell viability and apoptosis. Quantitative reverse transcription polymerase chain reaction was utilized to determine circ‐Ttc3 and miR‐449a expression. Western blot was performed to examine apoptosis‐associated proteins expression. Subsequently, the above‐described investigations were reperformed after miR‐449a upregulation. Results Hypoxia induced apoptosis and declined viability in HaCaT cells. Circ‐Ttc3 expression was enhanced after transfection with circ‐Ttc3 expressing vector. Overexpressing circ‐Ttc3 raised viability and reduced apoptosis in HaCaT cells. Moreover, miR‐449a expression was elevated by hypoxia and reversed by overexpressing circ‐Ttc3. And circ‐Ttc3 exerted its effect via downregulating miR‐449a. Finally, overexpressing circ‐Ttc3 activated nuclear factor kappa‐B (NF‐κB) and phosphatidylinositol 3 kinase (PI3K)/protein kinase B (AKT) pathways via downregulating miR‐449a. Conclusion Our data suggested that circ‐Ttc3 alleviated hypoxic injury and activated NF‐κB and PI3K/AKT pathways by downregulating miR‐449a in HaCaT cells.
