Circulating antibodies against c‐MYC oncogene product in sera of colorectal cancer patients

Abstract: We developed a Western blot assay using purified human c-myc protein expressed in E. coli in order to look for circulating anti-c-myc antibodies in human sera. The presence of IgG antibodies against c-myc was observed in 25 out of 44 sera from patients with colorectal cancer diagnosed and treated at the Hôpital Curie in Paris, compared to the sera of 46 normal donors of which only 8 samples were positive (p = 0.001).

“…These results indicate that ECPKA might be a cancer antigen. There is increasing evidence that patients with cancer produce autoantibodies against antigens in their tumors (12)(13)(14)(15), suggesting that such autoantibodies could have diagnostic/prognostic value. We speculated that ECPKA excretion might elicit the induction of serum autoantibodies and that the presence of such autoantibodies could serve for cancer detection.…”

“…Immunogenic tumor-associated antigens have been described primarily for malignant tumors including melanoma, breast cancer, colon cancer, ovarian cancer (Real et al, 1988;Ben-Mahrez et al, 1990;Gallagher et al, 1991;Disis et al, 1994;Disis and Cheever, 1996;Boon and Lloyd, 1997). However, dierent studies have indicated that meningiomas are also capable of inducing an immune response in the host.…”

“…For example, the identification of mutant ras genes by polymerase chain reaction and direct sequencing of DNA isolated from the serum or plasma of three patients with pancreatic cancer has been described (89 (96). Antibodies to the myc protein have also been described in the sera of patients with myeloid leukemia and lymphoma, including Burkitt's lymphoma (97).…”
