Class I MHC-Binding Characteristics of the 129/J Ly49 Repertoire

Abstract: The Ly49 family of NK cell receptors and its MHC-binding characteristics have only been well characterized in C57BL/6 (B6) mice. Previous studies have shown that 129/J mice express unique Ly49 genes that are not found in the B6 strain. Screening of a 129/J cDNA library led to the discovery of 10 distinct full-length Ly49-related coding sequences (Ly49e, g, i, o, p, r, s, t, u, and v). Although 129/J mice share identical class I MHC (Kb and Db) transcripts with B6 mice, only one Ly49 is identical in the two str… Show more

“…However, the region in disequilibrium with susceptibility extended over distally to Ly49d. These results together with the localization of Cmv1 as 117,120 with respect to genetic markers used for haplotype analysis. Results from two different reports 121,122 were compiled to construct the individual haplotypes.…”

“…In addition, the complex structural organization of the Ly49 gene family further complicated the analysis of the Cmv1 domain as many of the Ly49 genes have different copy numbers and genomic organization in inbred strains, making it difficult to distinguish allelic variants of the same or of distinct Ly49 gene copies. 117,120 Thus as an alternative to fine mapping, haplotype analysis of a set of loci linked to Cmv1 was applied to a panel of 18 inbred strains characterized by their pattern of resistance or susceptibility to MCMV. 118,119 A compilation of the results obtained for 27 independent markers evenly distributed over 1.8 Mb of genomic DNA demonstrated the presence of three major classes of independent haplotypes at this region (Figure 3).…”

“…However, careful analysis of the phenotype of these NK cells suggested that they carried the 129/J repertoire of Ly49s, not that of B6, mice (38). This suggested that our observed differences might be due, in part, to the genetic background of these mice.…”

“…The cDNA for Ly49r was cloned and has been reported elsewhere (38). Briefly, a cDNA library from IL-2-stimulated 129/J liver NK cells was constructed in the pBK-CMV vector (Stratagene, La Jolla, CA) by Cell and Molecular Technologies (Philipsburg, NJ).…”

“…However, it is possible that these mAbs bind to additional NOD Ly49 as seen for 129 NK cells. 29 Ly49 expression by splenic NKT cells of NOD mice was also evaluated. Relative to B6 NKT cells, very few NOD NKT cells expressed Ly49.…”
