Clearance of calcium pyrophosphate dihydrate crystals in vivo

Abstract: Synthetic triclinic calcium pyrophosphate dihydrate crystals were uniformly trace-labeled with Ytterbium-169 ('Tb), a pure gamma-emitting isotope with a halflife of 31 days. The solubility of the labeled crystals was similar to that of cold synthetic crystals. The clearance rate of labeled sterile crystals, sieved to obtain the desired size, was determined after injection of microgram quantities into 4 arthritic human and 3 normal adult rabbit joints and corrected by the observed rate of clearance of free ' T … Show more

“…In rabbits, intra-articular injection with autologous blood significantly decreases the clearance rate of calcium pyrophosphate crystals from knee joints 107. In vitro iron, as Fe 2+ but not as Fe 3+ ions, inhibits pyrophosphatase,108 leading to a diminished hydrolysation of inorganic pyrophosphate109 and as such contributes to the precipitation of inorganic pyrophosphate with calcium.…”

“…Chondrocalcinosis may be found in some patients and contribute to the degenerative joint processes. Indeed, iron can precipitate calcium pyrophosphate crystal formation in vitro 6. Also, iron can be found in the synovium of patients with HH at the time of joint replacement surgery.…”

“…Crystals were crushed in an agate mortar and sieved to yield 10 -20-m aggregates, which were sterilized and rendered pyrogen-free by heating at 200°C for 90 min. CPPD crystals were made by methods published earlier (22) and sieved, sterilized, and treated in a similar fashion as BCP crystals.…”
