Clinical Efficacy of Almitrine-Raubasine

Abstract: Different pharmacological properties of almitrine-raubasine show that this combination may be a good therapy for the treatment of age-related cerebral disorders and functional rehabilitation after stroke. Many clinical studies have been carried out in France and in the rest of Europe, confirming the value of this compound in such situations. Without discussing the complexity of clinical trials in both the areas of cognitive disorders and stroke, we shall present two studies demonstrating the beneficial effects… Show more

“…Heteroyohimbines are a prevalent subclass of the monoterpene indole alkaloids (Corynanthe type skeleton), having been isolated from many plant species, primarily from the Apocynaceae and Rubiaceae families 1 . These alkaloids exhibit numerous biological activities: ajmalicine is an α1-adrenergic receptor antagonist 2 3 4 5 , and mayumbine (19-epi-ajmalicine) is a ligand for the benzodiazepine receptor ( Fig. 1 ) 6 .…”

“…It is crucial to exercise caution when employing this combination until further clarification regarding the mechanism, timing, and dosage is obtained. Additionally, almitrine is frequently used in conjunction with another drug called raubasine, which acts as a vasodilator, for the treatment of age-related cerebral disorders ( Allain and Bentué-Ferrer, 1998 ) and certain pulmonary diseases. Notably, raubasine has demonstrated anti-SARS-CoV-2 effects in in vitro and animal studies, suggesting its therapeutic potential ( Kumar et al, 2021 ; Mohseni et al, 2022 ).…”

“…A double-blind controlled study involving patients with memory loss, lack of concentration, impaired mental alertness, and emotional instability supported that almitrine-raubasine could improve cognitive impairments [ 35 ]. Another controlled multicenter study investigating patients with cognitive decline (assessed by MMSE, SCAG) again suggested almitrine-raubasine significantly improved symptomatology compared with placebo [ 38 ]. Three other trails conducted in China involving 206 patients with vascular dementia also supported significant beneficial effect of almitrine-raubasine combination on the improvement of cognitive function measured by MMSE [ 39 ], although high risk of bias was observed.…”
