Far infrared ray (FIR) is non-ionizing electromagnetic radiation with wavelengths of 4-16 mm. Ceramic far infrared ray emitting materials (cFIR) are sources of FIR that exhibit only non-thermal effects at room temperature. Certain physical, chemical and biological effects of cFIR irradiation were investigated in this study that heretofore has not been well characterized. We demonstrated that cFIR irradiation reduced the size of water clusters, and significantly increased the freezing temperature of water. We also observed an increase in the volatility of a complex mixture of alcohol, water and solutes and recorded elevated total phenol in green tea infusions prepared using cFIR irradiated water. The effects of cFIR irradiated water on living cells were also investigated. The MC3T3-E1 murine osteoblast cells grown in the presence of cFIR irradiated water exhibited anti-oxidative effects on H 2 O 2 -mediated toxicity, as evidenced by late stage increase in intracellular alkaline phosphatase. The presence of cFIR irradiated water also resulted in significant decrease in COX-2 production in the chondrosarcoma cell line, SW1353, in response to lipopolysaccharide induction. Based on our findings, the possible anti-inflammatory effects of cFIR irradiated water and implications of our study with regard to bone and joint health were discussed.

Celecoxib has been effective in the prevention and treatment of chronic inflammatory disorders through inhibition of altered cyclooxygenase-2 (COX-2) pathways. Despite the benefits, continuous administration may increase risk of cardiovascular events. Understanding microbiome-drug-host interactions is fundamental for improving drug disposition and safety responses of colontargeted formulations, but little information is available on the bidirectional interaction between individual microbiomes and celecoxib. Here, we conducted in vitro batch incubations of human faecal microbiota to obtain a mechanistic proof-of-concept of the short-term impact of celecoxib on activity and composition of colon bacterial communities. Celecoxib-exposed microbiota shifted metabolic activity and community composition, whereas total transcriptionally active bacterial population was not significantly changed. Butyrate production decreased by 50% in a donor-dependent manner, suggesting that celecoxib impacts in vitro fermentation. Microbiota-derived acetate has been associated with inhibition of cancer markers and our results suggest uptake of acetate for bacterial functions when celecoxib was supplied, which potentially favoured bacterial competition for acetyl-CoA. We further assessed whether colon microbiota modulates anti-inflammatory efficacy of celecoxib using a simplified inflammation model, and a novel in vitro simulation of the enterohepatic metabolism. Celecoxib was responsible for only 5% of the variance in bacterial community composition but celecoxib-exposed microbiota preserved barrier function and decreased concentrations of IL-8 and CXCL16 in a donor-dependent manner in our two models simulating gut inflammatory milieu. Our results suggest that celecoxib-microbiome-host interactions may not only elicit adaptations in community composition but also in microbiota functionality, and these may need to be considered for guaranteeing efficient COX-2 inhibition.npj Biofilms and Microbiomes (2020) 6:9 ; https://doi.

Severe pain after third molar surgery is often encountered and more effective treatment regimes are warranted. The objective of this study was to evaluate if the combination of paracetamol and rofecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, improves analgesic effects following third molar surgery compared with rofecoxib alone. Paracetamol alone was also evaluated. Altogether 120 patients with moderate to severe pain after third molar surgery were given a single postoperative dose of one of the following treatments: rofecoxib + paracetamol; rofecoxib alone; paracetamol alone; or placebo. Patients assessed level of pain and pain relief every 30 min for 8 h after surgery, and made a global evaluation of the medication 4 and 8 h after surgery. Paracetamol and rofecoxib combined improved the analgesic effect compared with rofecoxib alone for the first 1.5 h. Rofecoxib alone and the combination of paracetamol and rofecoxib had a significantly better analgesic effect than paracetamol alone from 3 h onwards. The early onset of pain relief for the combination of paracetamol and rofecoxib, compared with rofecoxib alone, could be of great importance when treating acute pain after third molar surgery. After data collection for this study, rofecoxib was withdrawn from the market as a result of reported fatal cardiovascular events. Whether this is relevant for short-term use is unknown, but it has to be considered before rofecoxib may be used for pain relief following third molar surgery.
