Clinical significance of a single nucleotide polymorphism and allelic imbalance of matrix metalloproteinase-1 promoter region in prostate cancer

Abstract: Abstract. Matrix metalloproteinase-1 (MMP-1) is associated with cancer invasion and metastasis. The 2G allele of the polymorphic site in the MMP-1 promoter was demonstrated to have a higher transcription activity than the 1G allele. Allelic imbalance (AI) at 11q22 harboring the MMP-1 is frequently observed in various cancers and may be associated with an advanced disease. We conducted a case-control study to determine the association of the MMP-1 genotype with susceptibility to prostate cancer involving 283 pr… Show more

“…studies, several polymorphisms in the promoters of a number of MMP genes have been well characterized. These polymorphisms may affect the production or gene expression of these MMPs in an allele-specific manner (Piccoli et al, 2007;Tsuchiya et al, 2009;Wieczorek et al, 2013;Yaykas xli et al, 2014). Furthermore, there is an increasing evidence indicating that these functional polymorphisms may contribute to interindividual differences in a wide spectrum of cancer susceptibility (Decock et al, 2008;Pereira et al, 2012;Wieczorek et al, 2012).…”

“…Nine MMPs (MMP-1, -3, -7, -8, -10, -12, -13, -20, and -27) are clustered in the 11q22.3 region of chromosome 11 (Figure 2A). Chromosomal abnormalities and multiple polymorphisms in the 11q22.3 region predispose to prostate and lung cancer (57, 58) and coronary artery aneurysm formation in patients with Kawasaki’s disease (59). The approximately 0.43 Mb 11q22.3 region is largely deficient of CpGIs.…”
