Clock regulation of dietary lipid absorption

Abstract: Purpose of review-To summarize the new knowledge about the regulation of dietary lipid absorption by circadian locomotor output cycles kaput (Clock) and Nocturnin.Recent findings-Recent findings have shown that Clock and Nocturnin, proteins involved in circadian regulation, play an important role in the regulation of dietary lipid absorption. Clock deficiency increases, whereas Nocturnin deficiency decreases lipid absorption. Clock plays a role in turning off the genes involved in lipid absorption at the onset… Show more

“…Rhythmic triglyceride content was shown to be increased in Clock mutant mice via decreased interaction of Clock with small heterodimer partner ( Shp ), a nuclear receptor that represses Cyp7a1 gene expression, which resulted in sustained microsomal triglyceride transfer protein expression. 31 , 32 Liver gene expression of both Shp and microsomal triglyceride transfer protein were reduced after sleep disruption (data not shown), indicating differential gene expression changes in response to distinct types of circadian disruption (genetic or behavioral). Interestingly, Shp − / − mice are protected against fatty liver disease, and knockdown of the clock gene neuronal PAS domain protein 2 ( Npas2 ) reversed this phenotype and resulted in steatosis.…”

“…Night shift work is associated with both sleep deprivation and disrupted melatonin production (Burch JB et al, 2005) [40] and melatonin plays a key role in the circadian rhythms [41] . Two proteins involved in circadian regulation, CLOCK (which stands for circadian locomotor output cycles kaput) and nocturnin, play an important role in the regulation of dietary lipid absorption [42] . CLOCK plays a role in turning off the genes involved in lipid absorption at the onset of the day, and CLOCK deficiency increases lipid absorption.…”

“…The promoter region of Noc harbors an E-box, a cyclic AMP response element (CRE) and a photoreceptor-conserved element II (PCE II), all of which are under circadian control [52,56,57]. Noc is an important mediator of lipid metabolism, adipogenesis, glucose homeostasis, inflammation and osteogenesis [53,58]. In the sponge Suberites domuncula , Nocturnin regulates the deadenylation of the glycogenin mRNA, which encodes a key metabolic enzyme for carbohydrate/glycogen metabolism.…”
