“…In the present study, BA alone did not favor higher shoot proliferation, and MS or WPM devoid of growth regulators did not induce multiple shoot induction. Multiple shoot proliferation from cotyledonary node explants was successful in other tree species (Buendia-Gonzalez et al 2007;Nayak et al 2007).…”

“…The infl uence of cytokinins on shoot growth was examined by placing the explants on media enriched with adenine -BA or 2iP -or phenylurea -TDZ -derivatives (Table I). BA (6-benzylaminopurine) is one of the most commonly applied cytokinins, often used for microshoot induction in legume plants (Barik et al, 2007;Buendia-Gonzales et al, 2007), whereas the less popular 2iP has been shown to effectively induce morphogenesis in certain species (Luczkiewicz and Piotrowski, 2005;Maity et al, 2005). Of the auxin group, indole-3-butyric acid (IBA) was tested, as it has previously been shown to exhibit positive effects on shoot morphology (reduced vitrifi cation), especially when applied together with cytokinins (Rout et al, 2000).…”

“…Therefore, it was decided to subject the explants to further elongation. Unfortunately, the explants cultivated on the IBA-supplemented M10 medium for 60 d exhibited several unfavourable features, such as leaf loss, shoot tip withering, and extensive browning (Table II), probably as a result of the excessive osmotic potential of the growth medium (Rout et al, 2000;Gou et al, 2006;Buendia-Gonzales et al, 2007;Husain et al, 2007). The application of medium with reduced sucrose concentration -E1 -at the 1 st elongation stage yielded similar results (in terms of shoot length and morphology) as in the case of M10 medium (Fig.…”

“…), located around the village of Ejido Llanos de la Angostura in the Mexican State of San Luis Potosí. The seeds were scarified mechanically as described by Buendía-González et al (2007). The scarified seeds were washed in 5% (w/v) commercial detergent for 5 min, surface sterilized with 50% (v/v) ethanol for 5 min, and put into a 10% (w/v) sodium hypochlorite (solution \ 5% active Cl) for 5 min.…”
