Cloning and Characterization of a Gene Encoding an Immunoglobulin-Binding Receptor on the Cell Surface of Some Members of the FamilyTrypanosomatidae

Abstract: Several members of the Trypanosomatidae family, when freshly isolated from their mammalian hosts, have immunoglobulins adsorbed to their cell surfaces. However, a significant portion of these antibody molecules is not parasite specific, i.e., the immunoglobulins are bound to the parasite's cell surface molecules via noncognitive interactions. It has been proposed that this noncognitive adsorption of immunoglobulins to the parasite is mediated by an Fc-like receptor present in several members of the Trypanosoma… Show more

“…Polyclonal B cell activation (86,87) and parasite Fc-like receptors (88)(89)(90)(91) are thought to be partially responsible for anti-parasite ineffective humoral responses. In diverse infection models of strong polyclonal B cells responses, the most common isotypic distribution pattern is characterized by an increase in IgG2 subclasses (87,(92)(93)(94).…”

“…A rabbit anti-soluble lysate antigenic protein of L . infantum antiserum was prepared as we have previously described [ 24 ]. Briefly, 100μg of leishmanial proteins suspended in 1 ml of PBS were emulsified with 1 ml of incomplete Freünd’s adjuvant (IFA).…”

“…Supernatants from each of the 20 hybridoma clones were tested for their reactivity with synthetic purified 20mer peptides covering the entire full length of each biomarker and overlapping by 10 amino acids. Reactivity was tested by direct ELISA as previously described [31]. Peptides were synthesized by GenScript (Piscataway, NJ).…”
