Cloning and Sequencing of a Bottle-Nosed Dolphin (Tursiops truncatus) Interleukin-4-Encoding cDNA

Abstract: ABSTRACT. Using polymerase chain reaction (PCR), a bottle-nosed dolphin (Tursiops truncatus) interleukin-4 (IL4) cDNA was cloned and sequenced. IL4 specific primers were based on the 5' and 3' untranslated regions of the human and murine IL4 gene. The dolphin IL4 cDNA is 528 base pairs in length and contains an open reading frame of 402 nucleotides coding an IL4 precursor of 133 amino acids, with the putative signal peptide of 24 amino acids. Analysis of the mature amino acid sequence shows three potential N-l… Show more

“…The immune systems of terrestrial mammals, such as human and mouse are well understood, while those of marine mammals, living in the aquatic environment, are not. Little is known about cytokines in marine mammals with the exception of interleukin-2 (IL2) [15], interleukin-4 (IL4) [10] and interleukin-6 (IL6) [12]. In this study, the nucleotide and predicted amino acid sequences for the bottle-nosed dolphin IFN-γ are described and compared with those of other species.…”

“…Moreover, the dolphin interleukin-4 (IL4) gene sequence is also similar to the bovine one [10]. In the amino acid sequence level, thus, cetacean cytokines tend to resemble bovine ones.…”

“…The immune systems of terrestrial mammals such as human and mouse are well understood, while those of marine mammals, which encounter a wide variety of infectious agents in their environments are not. Little is known about cytokine genes in marine mammals with the exception of interleukin-2 (IL-2) [19], interleukin-4 (IL-4) [10], interleukin-6 (IL-6) [13] and interferon-γ (IFN-γ) [11]. In this study, the nucleotide and predicted amino acid sequences of the bottle-nosed dolphin IL-1α and IL-1β are described and compared with those of other species.…”

“…In addition, IL-4 of some ruminants like cow [24], African buffalo [25], sheep [26], and goat [6] was also previously cloned and identified. In addition, IL-4 has been identified and reported in chimpanzee [27] and bottle-nosed dolphin [28]. Wild animals are presumed to possess stronger immune system as compared to their domestic counterparts.…”
