Abstract:The objective of this study is to evaluate (in vitro and in vivo) seven plant extracts and their liquid fractions against four Penicillium italicum isolates. The in vitro study revealed that a concentration 520 µg/ml of crude extract of sticky fleabane leaves or cinnamon bark generated maximum percentage inhibition of 54% and 43%, respectively, against tested fungal isolates. A concentration of 130 µg /ml from each extract (except harmal and garlic where, 390 µg/ml were required) caused complete inhibition of fungal growth of isolates Pi.1 and Pi.3 infecting orange fruit. A concentration of 130 µg/ml of nightshade fruit, fenugreek or sticky fleabane extract inhibited completely the growth of isolates Pi.3 and Pi.5 infecting lemon fruits, whereas a concentration of 390 µg/ml was required to inhibit the growth of isolate Pi.1. Methanolic fractions of cinnamon, garlic or sticky fleabane completely inhibited the growth of fungal isolates. The IC 50 values for these fractions were found to be in the range of: 11.2-24; 30.25-31.50; 25.0-36.0 µg/ml, respectively.A concentration 20 µg/ml of cinnamon hexane fraction inhibited the growth of the fungal isolates, with IC 50 values of 13, 13.75, 14 and 13 µg/ml, respectively, obtained against isolates Pi.1, Pi.3, Pi.5 and Pi.6. The nightshade hexane fraction completely inhibited the growth of isolates Pi.1 and Pi.3 with IC 50 values of 80 and 37.5 µg/ml, respectively. Cinnamon aqueous fraction completely inhibited the growth of isolates Pi.1 and Pi.5 (IC 50 were 61.25 and 58.5 µg/ml, respectively).

Bruchid resistance is regulated by defensin gene. Vigna radiata plant defensin 1 (VrPDF1) inhibits alpha-amylase activities in insect gut; therefore, insect will die of undigestion starch. VrPDF1 content in mungbean seeds is very low. Hence an increase in content of VrPDF1 in mungbean seeds leads to enhance alpha -amylase inhibition in larvae and bruchids, which is necessary for the study to improve bruchid resistance in mungbeans. This article presents the results of VrPDF1 gene expression in T1 generation transgenic tobacco seeds. It was confirmed that VrPDF1 gene was attached to genome of tobacco plants and translated to synthesize VrPDF1. Recombinant VrPDF1 protein was successfully expressed in seeds of four transgenic tobacco lines (T1 -7, T1-8, T1-10, T1-11), among which T1-10 line had the highest recombinant VrPDF1 content, reaching 8.57 g mg -1 total protein. The extract containing recombinant VrPDF1 from the transgenic tobacco lines effectively inhibited the activity of alpha-amylase from the intestine of larvae and weevils. However, the protein extract solution from T1 -10 line had the strongest inhibitory effect, so the activity of alpha-amylase was only 18.89% compared to controls. The analysis results of VrPDF1 gene expression in transgenic tobacco plants are fundamental to the transfer of pPhaso-dest-VrPDF1 vector into mungbean to improve bruchid resistance of mungbean and contribute to improving mungbean preservation.

SynopsisPlant defensins are small cysteine-rich peptides and exhibit antimicrobial activity against a variety of both plant and human pathogens. Despite the broad inhibitory activity that plant defensins exhibit against different micro-organisms, little is known about their activity against protozoa. In a previous study, we isolated a plant defensin named PvD 1 from Phaseolus vulgaris (cv. Pérola) seeds, which was seen to be deleterious against different yeast cells and filamentous fungi. It exerted its effects by causing an increase in the endogenous production of ROS (reactive oxygen species) and NO (nitric oxide), plasma membrane permeabilization and the inhibition of medium acidification. In the present study, we investigated whether PvD 1 could act against the protozoan Leishmania amazonensis. Our results show that, besides inhibiting the proliferation of L. amazonensis promastigotes, the PvD 1 defensin was able to cause cytoplasmic fragmentation, formation of multiple cytoplasmic vacuoles and membrane permeabilization in the cells of this organism. Furthermore, we show, for the first time, that PvD 1 defensin was located within the L. amazonensis cells, suggesting the existence of a possible intracellular target.
