Cocaine increases immunoglobulin heavy chain binding protein and caspase-12 expression in the rat dorsal striatum

Abstract: These findings suggest that acute or repeated cocaine administration can cause ER stress response in the dorsal striatum in which NMDA and D1 dopamine receptors participate in the mediation of the process.

“…In conclusion, the stimulation of NMDA receptors in the shell of NAc and the effects of dopamine antagonists on presynaptic receptors might cause the increase in dopamine levels in the striatum and disrupt Ca 2+ homeostasis leading to the endoplasmic reticulum (ER) stress response [4,72]. In addition, stimulating dopamine and glutamate receptors increases nitric oxide efflux which activates PARP-1 in the central nervous system [69,86].…”

“…We also know from the literature that NAc dopamine and glutamate signalling interactions are crucially required in behavioural reinforcement and habit formation and dopamine can modulate excitatory glutamatergic projection from the PFC [82]. The elevation of extracellular dopamine and glutamate levels in the striatum might disrupt Ca 2+ homeostasis leading to the endoplasmic reticulum (ER) stress response [4,72]. The stimulation of dopamine D 1 receptors in the mouse neostriatum activates the cAMP/protein kinase A (PKA) pathway [76].…”

“…For instance, activation of NMDA receptors enhances an extracellular signal regulated kinase 1/2 (ERK1/2) signalling pathway, which results in cyclic AMP response element binding protein (CREB) phosphorylation and c-fos ex- pression in rat dorsal striatum (Choe & Wang 2001;Choe et al 2004). These data suggest that acute exposure to cocaine is able to modulate neuronal activity via activation of the NMDA receptor-regulated gene 2 isoform 2, which may be associated with psychomotor activity (Ahn et al 2007). Androgen receptor-related apoptosis-associated protein CBL27, isoform CRA b and seminal vesicle secretory protein 2 were altered by acute cocaine and are involved in sexual reproduction.…”

“…Increased levels of glutamate caused by repeated cocaine administration upregulate the endoplasmic reticulum stress protein and caspase-12 expression in the dorsal striatum (Shin et al 2007). Furthermore, N-methyl-D-aspartate (NMDA) receptors play an important role in the regulation of the protein expression (Ahn et al 2007). These findings suggest that repeated cocaine administration can lead to neuropharmcological, behavioral, or reinforcing responses in the dorsal striatum via NMDA receptor-coupled signaling cascades.…”
