Combined Effect of Bilateral Ovariectomy and Anterior Cruciate Ligament Transection With Medial Meniscectomy on the Development of Osteoarthritis Model

Abstract: ObjectiveTo investigate the combined effect of bilateral ovariectomy (OVX) and anterior cruciate ligament transection (ACLT) with medial meniscectomy (MM) on the development of osteoarthritis (OA).MethodsTwenty female 15-week-old Sprague-Dawley rats were used. Five rats in each group underwent bilateral OVX (OVX group), bilateral ACLT with MM (ACLT with MM group), bilateral OVX plus ACLT with MM (OVX plus ACLT with MM group), and sham surgery (SHAM group). All the rats were subjected to treadmill running for 4… Show more

“…Numerous animal models of OA have been developed to mimic pathologic human cartilage 22 . Among these different models, transection of the ACLT+MM has been one of the most widely used to induce OA, The joint instability caused by ACLT damage further leads to biological mechanics changes resemble the morphologic and biochemical changes seen in human OA 23 . ACLT+MM model provides progressive lesions that develop over time which is useful to study early stages of the OA 24 .…”

“…In this work we used only male rats. Current data on ACLT in female rats is limited; in females, ACLT is mostly used in combination with ovariectomy, which was not relevant to our study . Additional similar studies should be carried out in female rats to validate the relevance of our data.…”

“…The study was approved by the local Ethics Committee and performed in compliance with the Guidelines for the Care and Handling of Experimental Animals of the Laboratory Animal Center, Shanghai Tongji University. The study used the murine anterior cruciate ligament transection (ACLT)-induced model of osteoarthritis (OA) [ 16 ].…”

“…The experiments were repeated in triplicate, resulting in a total of 72 mice being used throughout the study. ACLT surgery was performed as previously described [ 16 ]. Briefly, the anterior cruciate ligament of the right knee was transected to generate a destabilized OA animal model.…”
