Combined therapy of multidrug-resistant human lung cancer with anti-P-glycoprotein antibody and monocyte chemoattractant protein-1 gene transduction: The possibility of immunological overcoming of multidrug resistance

Abstract: Lung cancer is the most common cause of cancer deaths in males and females. Although small-cell lung cancer is sensitive to chemotherapy, it gradually becomes resistant due to the emergence of multidrug-resistant cells. P-glycoprotein (P-gp) encoded by the mdr1 gene is one of the key molecules in MDR. It has been shown to bind anti-cancer drugs, to be an ATPase, and to be localized in the plasma membranes of MDR cells (Hamada and Tsuruo, 1988). Expression of the mdr1 gene was found to be elevated in intrinsica… Show more

“…MCP -1 has been reported to suppress growth and metastasis of multidrug -resistant lung cancer cells treated with the anti -P -glycoprotein antibody that induces antibody -dependent cellular cytotoxicity to the tumor cells in vivo. 46,47 In support of this notion, the treatment with both HSV-tk / GCV and MCP -1 was more effective in killing HCC cells than that with either HSV-tk / GCV or MCP -1. Thus, MCP -1 may be used to induce tumor regression of HCC even in combination with suicide gene delivery.…”

“…ADCC was measured using a standard 4 h 51 Crrelease assay (36,37). For blocking, labeled target cells were preincubated with the cmHsp70.1 mAb, the IgG1 isotype-matched control mAbs, SPA810 mAb, the theta-specific Ox7.11 (50 μg/mL, each), or the cmHsp70.1 Fab fragment (50 μg/mL).…”
