Comparative cytogenetic mapping of three genes involved in sex determination in four species of the family Canidae

Abstract: Advanced knowledge of the canine genome facilitates progress in studies on genome maps of other canids, including species considered also as farm animals. In this study canine BAC (Bacterial Artificial Chromosome) probes, harbouring three genes involved in sex determination (SOX9 -sex determining region Y-box 9, AMH -anti-Müllerian hormone and AR -androgen receptor), were mapped in the dog, red fox, arctic fox and Chinese raccoon dog chromosomes. Localization of these genes can be helpful in association studie… Show more

“…The coding region of the SRY gene and exons of SOX9 genes were amplified by polymerase chain reaction (PCR) by means of T personal thermocycler (Biometra, Gottingen, Germany). The sequences of primers were previously described in the studies of Bigliardi, Parma, and Peressotti () ( SRY ) and Nowacka, Nizanski, Klimowicz, Dzimira, and Switonski (), Nowacka‐Woszuk and Switonski () ( SOX9 ; Table ). PCR reaction for SRY gene consisted of initial denaturation at 95°C for 2 min followed by 35 cycles of denaturation at 95°C for 40 s, annealing at 56°C for 50 s and elongation at 72°C for 90 s. Final elongation was carried out at 72°C for 5 min.…”
