Comparative Proteomic Analysis of Rice Shoots Exposed to High Arsenate

Abstract: Consumption of arsenic contaminated water and cereals is a serious threat to humans all over the world. Rice (Oryza sativa "Nipponbare"), as a main cereal crop, can accumulate arsenic more than 10-fold that of in other cereals. To gain a comprehensive understanding of the response of rice subjected to 100 µM arsenate stress, a comparative proteomic analysis of rice shoots in combination with morphological and biochemical investigations have been performed in this study. The results demonstrated that arsenate s… Show more

“…Proteins were extracted using the phenol method according to previously description ( Liu et al, 2013 ). Briefly, the samples were ground to power with liquid nitrogen, homogenized in pre-cooled homogenization buffer (20 mM Tris/HCl (pH 7.5), 250 mM sucrose,10 mM ethylene glycol-bis(b-aminoethylether)-N,N,N′,N′-tetraacetic acid (EGTA), 1 mM PMSF, 1 mM DTT, and 1% Triton X-100), followed by centrifugation at 12 000 g for 20 min at 4°C.…”

“…Arsenate (AsV) induced NADP-dependent malic enzyme (NADP-ME), aspartate aminotransferase, NAD-dependent formate dehydrogenase (FDH), GAPDH and ATP-dependent protease, proteolytic subunit ClpP-like protein have been reported in rice leaves 19 . In another study, AsV stress altered the chloroplast 23 kDa polypeptide of photosystem II, Chloroplastic aldolase and Photosystem II stability/assembly factor HCF136 that leads to reduced photosynthetic rate and distorted chloroplast structure in rice leaves 20 . Studies on nutritional aspects vis-à-vis As have been targeted only to nitrogen 21 and phosphorus supplementations 22 .…”

“…AsV, though not a redox metalloid, can provoke cellular disruption in plants through the induction of oxidative stress caused by the generation of reactive oxygen species (ROS) during the conversion of AsV to As(III) (Gomes et al, 2012;Liu et al, 2013), while Cd, a redox metal, does not participate directly in cellular redox reactions (Clemens, 2006;Garg and Bhandari, 2014). Exposure to Cd drives oxidative injuries, such as lipid peroxidation, which leads to alteration in the membrane functionality and protein carbonylation (Schützendübel et al, 2001;Romero-Puertas et al, 2002;DalCorso et al, 2008DalCorso et al, , 2010Garg and Aggarwal, 2012).…”
