Structural changes in endosperm cells of germinating castor beans were examined and complemented with a cytochemical analysis of staining with diaminobenzidine (DAB) . Deposition of oxidized DAB occurred only in microbodies due to the presence of catalase, and in cell walls associated with peroxidase activity . Seedling development paralleled the disappearance of spherosomes (lipid bodies) and matrix of aleurone grains in endosperm cells . 6 to 7 days after germination, a cross-section through the endosperm contained cells in all stages of development and senescence beginning at the seed coat and progressing inward to the cotyledons . Part of this aging process involved vacuole formation by fusion of aleurone grain membranes . This coincided with an increase in microbodies (glyoxsomes), mitochondria, plastids with an elaborate tubular network, and the formation of a new protein body referred to as a dilated cisterna, which is structurally and biochemically distinct from microbodies although both apparently develop from rough endoplasmic reticulum (ER) . In vacuolate cells microbodies are the most numerous organelle and are intimately associated with spherosomes and dilated cisternae . This phenomenon is discussed in relation to the biochemical activities of these organelles. Turnover of microbodies involves sequestration into autophagic vacuoles as intact organelles which still retain catalase activity . Crystalloids present in microbodies develop by condensation of matrix protein and are the principal site of catalase formerly in the matrix .

By a combination of Sephadex gel filtration, CM-C and DEAE-C chromatography, 2 highly purified HCG fractions (dA: 18,000 IU/mg, dB: 20,000 IU/mg) were obtained from a crude preparation (2130 IU/mg) which was extracted from the urine of normal pregnant women in the first trimester. They were characterized by histological response of the ovary of hypophysectomized immature rats, bioassay (the Steelman-Pohley method and the OAAD method), amino acid analysis and determination of carbohydrate content, and their high homogeneity was proved by polyacrylamide gel electrophoresis, immunodiffusion and immunoelectrophoresis. The components confirmed were 17 amino acids, galactose, mannose, hexosamine and sialic acid. LH-like activity was dominant in fraction dA, and this fraction was richer in proline and valine than fraction dB and poorer in aspartic acid, glutamic acid, glycine, alanine, lysine and histidine. Fraction dB had dominant FSH-like activity in its biological character and was richer in acidic amino acids and sialic acid. Biological potency was particularly well correlated between LH-like activity of dA and hexose, and between FSH-like activity of dB and acidic amino acids and sialic acid. These results suggested that HCG is composed of 2 components. One has LH-like activity and the other has FSH-like activity. The ratio of these activities in HCG preparations differs according to the stage of pregnancy and biological and physicochemical properties of this hormone change throughout pregnancy. Ahstrrrci. The authors studied the secretion patterns of three chorionic polypeptide hormones (HCG. HCT and HCS) and the clinical usefulness of the assay of these hormones in patients with hydatidiform mole, by means of systematic serial estimation. in comparison with the secretion patterns in normal pregnancy. The serum levels of HCG and HCT in molar pregnancy were usually higher than in normal pregnancy but HCS levels were below the lower limit of normal pregnancy of the same gestational age. This dissociation between the chorionic glycoprotein hormones (HCG and HCT) and the simple protein hormone (HCS) is thought to be one of the characteristic findings in molar pregnancy. Although the amounts of HCG and HCT did not change 6 hours after the evacuation of hydatidiform mole, the concentration of HCS decreased rapidly and 60 min later it became undetectable. Thus, these polypeptide hormones disappeared in the order HCS, HCT and HCG after the evacuation of mole.
