“…Both strains showed a similar growth profile (Fig 2A). We observed that the MHY1-deletion strain accumulated somewhat more lipid than the wild type in both nitrogen sources tested (Fig 2B), in line with previous findings [25,26], further strengthening the beneficial traits of the MHY1-deletion. We also studied the fatty acid composition of the two strains, which did not show any significant changes (Fig 2C, detailed in supplement Table 1).…”

“…Finally, we tested the deletion of the MHY1 gene, which has a C2H2-zinc finger, DNAbinding domain on the C-terminal. Deletion of MHY1 has been shown to abolish hyphae formation [11,[24][25][26], while overexpression increased hyphae formation [11]. In our hands, the MHY1-deletion (OKYL029) was the only strain which abolished hyphae formation under all tested conditions (Fig 1).…”

“…An RNA-seq library was constructed using the NEBNext ® Ultra ™ RNA Library Prep kit for Illumina ® (New England Biolabs, Ipswich, MA, USA) according to the manufacturer's instructions. RNA library quality confirmation and RNA sequencing were carried out according to the process described by Wang et al (2018a).…”

“…In subsequent work, the same researchers showed that leucine biosynthesis was particularly downregulated when the yeast was grown under nitrogen-limited conditions, concomitantly with lipid accumulation ( Kerkhoven et al, 2017 ). A recent study looked at transcriptional changes in Y. lipolytica during lipid biosynthesis in strain carrying a MHY1 gene inactivation ( Wang et al, 2018 ); MHY1 encodes a C 2 H 2 -type zinc finger protein. They found that nearly 25% of annotated Y. lipolytica genes were expressed at significantly different levels, suggesting Mhyp plays a crucial regulatory role in various biological processes, including lipid and amino acid metabolism (again underscoring the interaction between these pathways).…”
