l-3,4-dihydroxyphenylalanine (l-DOPA) is a medically relevant compound in Parkinson’s disease therapy. Several extraction methods of l-DOPA from beans, including velvet and faba beans, have been described in the literature. However, these methods require the use of strong acids, long extraction times, or complex downstream processing, which makes the extraction of l-DOPA expensive and energy-demanding, limiting its industrial application. In addition, the stability of l-DOPA during the extraction process is critical, further complicating the extraction of adequate amounts of this amino acid. This work is the first report on a simple, rapid, greener, and robust extraction method of l-DOPA. The developed method consists of a quick homogenization step followed by a double extraction with 0.2% v/v acetic acid for 20 min and was applied to faba bean at a ratio of 1:25 with respect to the extracting solvent. This study also investigated the stability of l-DOPA during extraction and thermal treatment. The proposed method demonstrated to be robust and extraordinarily efficient for numerous cultivars of faba bean, velvet bean, and food products containing faba beans.

The total phenolic and flavonoid contents (TPC and TFC) from the genus Hosta with antioxidant and α-glucosidase inhibitory activities were reported for the first time. Sixteen extracts from the aboveground and underground parts of the four Hosta species, including H. plantaginea, H. ventricosa, H. ensata, and H. albofarinosa, using reflux extraction (RE) and ultrasound-assisted extraction (UAE) techniques have high TPC and TFC with good antioxidant and α-glucosidase inhibitory activities. Furthermore, no significant differences on extraction yields, TPC, and TFC were found between RE and UAE techniques. Additionally, extracts from the aboveground parts of the four Hosta species had higher TPC, TFC, antioxidant, and α-glucosidase inhibitory activities compared to the underground parts by means of RE or UAE techniques. Lastly, the extracts of H. albo-marginata displayed a very remarkable α-glucosidase inhibitory activity compared to the positive control acarbose. The relationships of sixteen extracts of the four Hosta species were analyzed by RE and UAE techniques between extraction yields, TPC, TFC, antioxidant activity, and α-glucosidase inhibitory activity. The present study demonstrated that H. plantaginea, H. ventricosa, H. ensata, and H. albofarinosa could be new sources of natural antioxidants and antidiabetes for pharmaceutical and industrial purposes.

Chemical Characterization of Constituents Isolated from Hamelia patens and Investigating Its Cytotoxic Activity

S RB cytotoxicity assay was performed for different extracts of Hamelia patens Jacq. family Rubiaceae; crude flowers (CF), crude leaves (CL), chloroform (Chl.L) and methanol (Me.L) fractions from leaves against liver (HepG-2) and breast (MCF-7) human carcinoma cell lines. Results demonstrated potent cytotoxic action against HEPG-2 for CF, Chl.L and Me.L with IC 50 = 47, 30 and 44.4 µg/mL, respectively. On the other hand, CF, CL, Chl.L and Me.L had exerted powerful effect against MCF-7 with IC 50 = 23.8, 25.5, 17.7 and 64 µg/mL, respectively. Chromatographic investigation of Me.L , resulted in the isolation and spectroscopic identification of rutin (1), isoquercetin (2) and soyasaponin Bb (3), which was isolated for the first time from the plant leaves and was identified by UPLC/ITMS/MS analysis. Comparative study for using ordinary method versus green methods of extraction was also inspected.
