Abstract: Compared with mixed marrow stromal cells, cloned osteoprogenitor cells can produce a larger amount of mature osseous tissue at an earlier time point during spine fusion.

“…Moreover, previously published rat spinal fusion studies assessing autologous bone grafting were observed to yield an inferior fusion rate when compared with hPSC treatment (0% fusion after 4 weeks and 11% fusion after 8 weeks with autologous bone grafting, compared with 80%-100% fusion in 4 weeks with hPSC treatment) [46,47]. MSCs derived from multiple sources such as bone marrow and adipose tissue have attracted large interest for replacement of autogenous bone in spinal fusion procedures [48][49][50][51][52]. BMSCs, however, are limited in supply, and it is estimated that only 1 BMSC can be isolated per 100,000 cells [53].…”

“…BMSCs, however, are limited in supply, and it is estimated that only 1 BMSC can be isolated per 100,000 cells [53]. To circumvent the shortage of MSCs, readily isolatable methods such as selective cell retention [54] and ex vivo cell culture methods have been developed [48][49][50]. Selective cell retention of osteoprogenitors are clinically available using Cellect Graft Preparation system (Dupuy, Raynham, MA, http://www.depuy.com); however, no clinical reports of success have been published [55].…”

“…Mesenchymal stem cells have been used in multiple tissue-engineering applications, and they differentiate into multiple lineages, including osteoblasts, chondrocytes, adipocytes, and muscle/tendon forming cells [9][10][11]23]. The multipotent, immortalized BMSC line used here is primarily osteogenic [24] and has been used in models of spinal fusion [19] and in the femur [22]. These cells also differentiate into adipocytes under the influence of steroids [20,21] or alcohol [22].…”

“…Bone marrow stem cells could serve as graft material and a vehicle to deliver therapeutic factors that enhance bone repair [9,13]. The efficacy of bone marrow implantation in the osteonecrotic zone was studied in prospective trials in humans and showed progression of the disease was avoided in nine of the 10 hips that received bone marrow implantation, whereas five of the eight hips in the control group had deteriorated to a late stage of the disease [14][15][16].…”
