Comparison of the Antidepressant-Like Effects of Estradiol and That of Selective Serotonin Reuptake Inhibitors in Middle-Aged Ovariectomized Rats

Abstract: This study investigated the effect of age and that of the post-ovariectomy (OVX) time interval on the antidepressant (AD)-like effects of estradiol (E2) and selective serotonin reuptake inhibitors (SSRIs) in middle-aged (10 month) OVX rats (10m-OVX). Acute or chronic effects of these treatments in 10m-OVX were compared with those (1) in young adult (4-month) OVX rats (4m-OVX) or with older (14-month) OVX rats (14m-OVX), at a short time: 2 weeks post-OVX (+2w) and (2) in 10m-OVX rats after a longer times: 4 or … Show more

“…Consistent with these discoveries, previous studies reported a poorer response of postmenopausal women to antidepressant treatments than premenopausal women [34,35]. The findings from this investigation were contrary to the results reported by Benmansour et al [36] who showed that the antidepressant-like effects of SSRIs were not affected by the length of hormone depletion. The inconsistency in these conclusions may be due to the difference in the initiation time of SSRI treatment in relation to OVX onset.…”

“…The inconsistency in these conclusions may be due to the difference in the initiation time of SSRI treatment in relation to OVX onset. Benmansour et al (36) studied the impact of length of hormone depletion by measuring the effects of SSRIs at 2 weeks, 4 months, or 8 months post-OVX. Based on accumulating evidence, EPO/EPOR and their downstream signaling pathways are potential targets in the treatment of depression [12].…”

“…The finding that QBA increases collagen production in fibroblasts [ 41 ] suggests an interaction with the estradiol system, as estradiol has the same effect [ 42 ]. Estradiol also increases neurite outgrowth [ 43 ], protects neurons from stress-related death [ 44 ], improves mood in a dose-dependent manner [ 45 , 46 , 47 , 48 ], and activates erk through non-genomic mechanisms [ 23 ]. Royal jelly has activity at estradiol receptors, yet its estrogenic activity may not cross the blood-brain barrier, as evidenced by RJ’s ability to induce vascular endothelial growth factor (VEGF) in the uterus but not the brain.…”

“…While luvoxamine reduces the clearance of 5-HT on rats about 10 months aged with 2 weeks, 4 and 8 months postovariectomy the same way as the 4 months rats [51]. Additionally, they detected that 5 μg/day of estradiol for 2 weeks subcutaneously via implantation of osmotic minipumps produces antidepressant efects on forced swim test on 4-month aged rats but not in 10-month aged rats both with 2 weeks of postovariectomy.…”

“…In this study, it is concluded that the age inluences the potency of estradiol on the 5-HTT, but its efects were strongly reduced if the period of postovariectomy is longer. On the other hand, the treatment with sertraline inhibited the 5-HTT and produced antidepressant-like efects without afected either by age or length of hormonal depletion [51].…”
