Comparison of the Cisterna Maturation-Progression Model with the Kiss-and-Run Model of Intra-Golgi Transport: Role of Cisternal Pores and Cargo Domains

Abstract: The Golgi complex is the central station of the secretory pathway. Knowledge about the mechanisms of intra-Golgi transport is inconsistent. Here, we compared the explanatory power of the cisterna maturation-progression model and the kiss-and-run model. During intra-Golgi transport, conventional cargoes undergo concentration and form cisternal distensions or distinct membrane domains that contain only one membrane cargo. These domains and distension are separated from the rest of the Golgi cisternae by rows of … Show more

“…2 of (Hudoklin et al 2009 )). This situation is similar to the situation in neurons, where the cis -most cisterna joins the Golgi stacks and is the opposite to the situation in fibroblasts, where the trans -most cisterna joins the Golgi stack (Mironov et al 2017 ; Beznoussenko et al 2022 ). However, it seems that the Golgi complex contains only limited amount of UPs.…”

“…This suggests that COPI vesicles are not involved in the transport of UP particles through the Golgi complex. On the other hand, it is known that COPI vesicles do not participate in the recycling of Golgi resident proteins (Mironov and Beznoussenko 2019 ; Beznoussenko et al 2022 ) and UP particles are rather big for diffusion along the membrane continuum within Golgi cisternae.…”

“…Samples were taken from 106 rats, namely, 25 normal male WKY rats subjected to standard cryoinjury, 55 male WKY and SHR rats subjected to other more complicated experiments, and 26 normal male WKY rats subjected to correlative light electron microscopy and immuno-electron microscopy labeling. Methodology and the details of all ethics rules are described in [ 50 , 51 , 52 ]. Briefly, all the experimental animal procedures were approved by the Committees of the Ivanovo State Medical Academy.…”

“…Brain cortex was excised and processed for EM examination and EM tomography as previously described (Beznoussenko et al, 2015). For immuno-labeling, the grids were prepared as previously described (Beznoussenko et al, 2022). Briefly, the grids were incubated for 10 min in 0.5% BSA-c in PBS (bovine serum albumin-c TM , acetylated, 10% in water; Aurion) to prevent nonspecific labeling, and then incubated on 10 μL droplets of primary anti-Gpr126 (ab218046; Abcam) diluted 1:10 for 2 h, and then with protein-A gold 10 nm (PAG10, CMC, Utrecht, The Netherlands) 1:50 diluted in blocking solution for 20 min at room temperature.…”
