Comparison of the determination of chloramphenicol residues in aquaculture tissues by time-resolved fluoroimmunoassay and with liquid chromatography and tandem mass spectrometry

Abstract: A competitive time-resolved fluoroimmunoassay (TR-FIA) was developed for the determination of chloramphenicol (CAP) residues in aquaculture tissues based on monoclonal antibodies. The limits of detection (LOD) were determined to be 0.04 ng g (1 and the limits of quantification (LOQ) were less than 0.15 ng g (1. The intra-assay variations were below 10% and the interassay variations ranged between 9.7 and 13.3%. The mean recoveries established at six concentration levels varied from 83.7 Á109.6% and the coeffic… Show more

“…Considering that, it is necessary to establish a rapid, sensitive assay for the determination of CAP, TAP and FF residues in food samples. A number of analytical methods have been developed to detect CAP, TAP and FF; these include high-performance liquid chromatography (Xie et al, 2011), gas chromatographymass spectrometry (GC-MS) (Shen et al, 2009), liquid chromatography tandem mass spectrometry (LC-MS/MS) (Barreto, Ribeiro, Hoff, & Dalla Costa, 2016;Li, Hu, Huo, & Xu, 2006;Rezende, Filho, & Rocha, 2012), enzyme-linked immunosorbent assay (Fodey, Murilla, Cannavan, & Elliott, 2007;Liu et al, 2014; and lateral flow immunoassay (LFA) (Bai et al, 2013;Guo et al, 2015;Samsonova, Cannavan, & Elliott, 2012;Xu, Xu, Ma, Liu, et al, 2015). Compared with chromatographic methods, immunoassays are simple, rapid and do not require expensive equipment and professional operator.…”

“…Therefore, the test strip and ic-ELISA had similar results, and the strip test results were closer to the actual addition amount. Our findings showed that the fluorescent immunoassay strip is reliable for the detection of Cd rice (Li et al, 2007). Therefore, the fluorescent immunoassay strip, which is inexpensive, fast, and does not require large instruments, can be used in the detection of Cd in rice, meeting the current demand for a large number of rapid detection of rice in the field (Arai et al, 2016).…”

“…Based on cell selection, the cells were subcloned three times. After several screenings, the optimum cell line was expanded and injected into mice by intraperitoneal injection (Li, Hu, Huo, & Xu, 2007). Extracted ascites were purified by the ammonium caprylate sulfate method (Li, Liu, Song, & Kuang, 2017), and the concentration was determined using NanoDrop One (Thermo Fisher Scientific, Shanghai, China).…”

“…There are many methods for detecting organophosphorus pesticides, such as gas chromatography (GC) (Xin, Qiao, Xu, & Zhou, 2013;Liu, Yang, & Kang, 2008), high-performance liquid chromatography (He, Luo, Jiang, & Qu, 2010;Li, Zhang, Kong, Qiao, & Xu, 2017), gas or liquid chromatography coupled with tandem mass spectrometry (Li, Hu, Huo, & Xu, 2006;Chung & Chan, 2010;Wang, Zhong, Mao, & Wan, 2015), and capillary electrophoresis (Chen & Fung, 2010). Most of these described methods offer low detection limits and good sensitivities but need expensive instruments and long analysis times.…”
