Comparison of the pharmacokinetics of metronidazole in healthy female volunteers following either a single oral or intravenous dose.

Abstract: 1 The plasma pharmacokinetics of metronidazole following a single dose (500 mg) of metronidazole have been investigated in a crossover study in healthy female volunteers, using assays specific for metronidazole and its metabolites 1-(2-hydroxyethyl)-2-hydroxymethyl-5-nitroimidazole (metabolite I) and 2-methyl-5-nitroimidazole-l-acetic acid (metabolite II). 2 No systematic differences, which could be related to the route of metronidazole administration, were observed in the area under the plasma metronidazole c… Show more

“…The biological half-lives of metronidazole found in this study agree well with the results of Houghton et al (7,8) and also agree well with the data presented by Welling and Monro (24). The half-lives determined with bioassays (10,14,17) were higher, presumably due to the biological activity of the hydroxy metabolite (18).…”

“…The half-lives determined with bioassays (10,14,17) were higher, presumably due to the biological activity of the hydroxy metabolite (18). The serum levels of this metabolite described above agree well with earlier observations (7,8). The acetic acid metabolite was detected in serum during the first 8 h after infusion at very low levels.…”

“…The predicted peak plasma metronidazole concentration at 'steady-state' during oncedaily dosing with benzoylmetronidazole equivalent to 2 g metronidazole was 21 ,g/ml. This maximum value was similar to that observed immediately after an infusion of 500 mg metronidazole (Houghton et al, 1979b). The mean value for the minimum metronidazole concentration was predicted to be equal to the minimum inhibitory concentration for most susceptible anaerobes (6.2 ,ug/ml ;Finegold & Sutter, 1972).…”

“…Additionally, data from nine healthy volunteers (Houghton et al, 1979a) are superimposed on each graph for comparative purposes. All pharmacokinetic parameters were calculated from individual plasma concentration or urinary data and mean results (± s.d.…”
