Compartmental Genomics in Living Cells Revealed by Single-Cell Nanobiopsy

Abstract: The ability to study the molecular biology of living single cells in heterogeneous cell populations is essential for next generation analysis of cellular circuitry and function. Here, we developed a single-cell nanobiopsy platform based on scanning ion conductance microscopy (SICM) for continuous sampling of intracellular content from individual cells. The nanobiopsy platform uses electrowetting within a nanopipette to extract cellular material from living cells with minimal disruption of the cellular milieu. … Show more

“…Due to their high-aspect ratio geometry and exceptionally sharp tips, nanopipettes offer an important advantage over conventional nanopores as they can be easily adapted for use in single cell interrogation 15,16 and as a macroscopic delivery vehicle for intracellular injection. [17][18][19][20][21][22] . To date, delivery of molecules has so far only been quantified in close proximity to the tip by using fluorescence spectroscopy.…”

“…22 Additionally, nanopipette probes still hold great promise as intracellular biosensors [23][24][25] and as tools for cell manipulation. [26][27][28] We recently developed a method to fabricate multifunctional nanoprobes starting from double-barrel quartz nanopipettes. Pyrolytic carbon is selectively deposited on one barrel while leaving the other one unchanged.…”

“…As well as being able to deliver material to a surface, the SICM nanopipette also allows singlecell nano-biopsies to be performed, through the extraction of a small amount of material from inside a cell which is then further analysed [83,84]. This material can be, for example, mRNA, the levels of which can be quantified using PCR.…”
