Complete Genome Sequence of Corynebacterium pseudotuberculosis I19, a Strain Isolated from a Cow in Israel with Bovine Mastitis

Abstract: This work reports the completion and annotation of the genome sequence of Corynebacterium pseudotuberculosis I19, isolated from an Israeli dairy cow with severe clinical mastitis. To present the whole-genome sequence, a de novo assembly approach using 33 million short (25-bp) mate-paired SOLiD reads only was applied. Furthermore, the automatic, functional, and manual annotations were attained with the use of several algorithms in a multistep process.

“…(27,37,57,94,113,149,166,224,267,327). As a result, large numbers of genomes of Corynebacterium species and strains, including C. aurimucosum, C. diphtheriae, C. efficiens, C. glutamicum, C. jeikeium, C. kroppenstedtii, C. pseudotuberculosis, C. ulcerans, and C. urealyticum, have been sequenced, and many are in the process of being sequenced (39,159,213,267,298,299,304,339). The family Corynebacteriaceae contains two genera, Corynebacterium and Turicella (57,103,187,191).…”

“…The orientation and ordering of these contigs was performed using BLAST (blastN) software to map the contigs against the FRC41 reference genome (8). Therefore, the BLAST results, the multi-FASTA files containing the contigs, and the FASTA files that contained the reference sequence were entered into the Graphical Contig Analyzer for All Sequencing Platforms software (G4ALL; http://g4all.sourceforge.net/) to manually curate and extend the contigs and generate the genome scaffold with gaps.…”
