Complete nucleotide sequence of tobacco streak virus RNA 3

Abstract: Double-stranded cDNA of in vitro polyadenylated tobacco streak virus (TSV) RNA 3 has been cloned and sequenced. The complete primary structure of 2,205 nucleotides reveals two open reading frames flanked by a leader sequence of 210 bases, an intercistronic region of 123 nucleotides and a 3'-extracistronic sequence of 288 nucleotides. The 5'-terminal open reading frame codes for a Mr 31,742 protein, which probably corresponds to the only in vitro translation product of TSV RNA 3. The 3'-terminal coding region p… Show more

“…No other ORFs longer than 200 nt were found on either plus or minus strands. Thus, the overall organization of PNRSV RNA3 is very similar to that of the RNAs 3 of PDV (Bachman et al, 1994), TSV (Cornelissen et al, 1984) al., 1983), brome mosaic virus (BMV) (Ahlquist et al, 1981) and cucumber mosaic virus (CMV) (Davies & Symons, 1988).…”

“…P3a is translated directly from RNA3, whereas the CP is translated from a subgenomic mRNA, RNA4, which is also encapsidated. The complete nucleotide sequences of the RNA3 of tobacco streak virus (TSV) (Cornelissen et al, 1984) and prune dwarf virus (PDV) (Bachman et aL, 1994) are available and the sequence data indicate that the RNA3 of ilarviruses share a genome organization similar to that of other members of the Bromoviridae.…”

“…To date only sequence data for the type member of the group (tobacco streak ilarvirus-TSV) has been published, although sequence data for other ilarviruses exist (Alrefai et al, 1994;Bachman et al, 1994). The full-length sequence for RNA-3 of TSV has been determined (Cornelissen et al, 1984) and data for approx. 200 nucleotides at the 3' end of RNA-2 are also known (Koper-Zwarthoff & Bol.…”

“…There was no deletion and addition between nucleotide sequences in the group. Cornelissen et al (1984) cloned and sequenced TSV RNA3 genome reveals that, complete sequence of 2,205 nucleotides of TSV RNA 3, confirming 140 bp 3'-terminal residues. Two long open reading frames starting with a methionine codon are revealed by this sequence.…”
