Comprehensive Analysis of the Clinical and Biological Significances of Endoplasmic Reticulum Stress in Diffuse Gliomas

Abstract: BackgroundAs a critical organelle for protein and lipid synthesis, the dysfunction of endoplasmic reticulum has a significant impact on multiple biological processes of cells. Thus, in this study, we constructed an ER stress-related risk signature to investigate the functional roles of ER stress in gliomas.MethodsA total of 626 samples from TCGA RNA-seq dataset (training cohort) and 310 samples from CGGA RNA-seq dataset (validation cohort) were enrolled in this study. Clinical information and genomic profiles … Show more

“…We downloaded the RNA sequencing data, gene mutation data and corresponding clinical information of bladder cancer from TCGA database 1 , and extracted an ERS-related genes from published literature ( 18 ). On the premise that correlation coefficient |R 2 | > 0.3 and p < 0.05 were considered to be statistically significant, we identified ERS-related lncRNAs through Pearson correlation analysis.…”

“…In Figure 3B , we can find that the curve is the flattest when k = 2 (u1 = 0.2, u2 = 0.8). So, the ideal number of clusters (k = 2) was found using CDF curves and consensus matrices, with patients being divided into two stable clusters ( Huang et al, 2021 ) ( Figure 3B ). Of the 406 BLCA samples, cluster 1 included 333 samples and cluster 2 included 73 samples.…”

“…After removing overlapping genes, the obtained ER stress-related gene collection had 272 genes. Among these, 256 genes were discovered in the TCGA dataset ( Huang et al, 2021 ) ( Supplementary Table S1 ), which were screened in this study using differential expression and univariate analysis. The analyses revealed 23 key ER-related genes in BLCA.…”

“…TIMER2.0 (Tumor Immune Estimation Resource 2.0, , accessed on 1 March 2022) [ 19 , 20 , 21 , 22 ] is a comprehensive resource for the systematic analysis of immune infiltrates across diverse cancer types. In the outcome module, the Cox proportional hazard model covariates were overall survival and FDCSP expression, and the normalized coefficient of the infiltrate for each model across HPV status in HNSC cancer types is presented in a heatmap.…”
