Concentration of SARS-CoV-2-Infected Cell Culture Supernatants for Detection of Virus-like Particles by Scanning Electron Microscopy

Abstract: There is currently a need for new rapid viral diagnostic electron microscopy methods. Although the gold standard remains the transmission electron microscopy (TEM) negative staining method for electron microscopic examination of samples containing a virus, difficulties can arise when the virus particle content of the sample that has to be examined is poor. Such samples include supernatants of virus-infected cells that can be difficult to examine, as sometimes only a few virus particles are released in the cult… Show more

“…Kataoka et al examined the morphological and distribution changes of influenza A virus particles in host cells by using transmission electron microscopy (TEM) and scanning electron microscopy (SEM) methods . As a powerful tool, electron microscopy was also applied to detect and describe the emerging virus. , Electron microscope-based detection has the advantages of simplicity, directivity, and high resolution. However, the method requires expensive instruments and professional operators and is restricted by low sensitivity and susceptibility to matrix interference.…”

“…Viruses in water can be concentrated based on their surface charge, using flocculation-precipitation methods or negative/positive membrane filters [ 51 , 52 ]. Size-based techniques can be used for viral concentration in both water and cell culture supernatants [ 53 , 54 ]. Selection of cell-free DNA (cfDNA) in plasma based on size can enrich viral DNA, as cfDNA size varies according to the species [ 55 ].…”
