Congenital microcephaly unrelated to flavivirus exposure in coastal Kenya

Abstract: Background: Zika virus (ZIKV) was first discovered in East Africa in 1947.  ZIKV has caused microcephaly in the Americas, but it is not known whether ZIKV is a cause of microcephaly in East Africa. Methods: We used surveillance data from 11,061 live births at Kilifi County Hospital in coastal Kenya between January 2012 and October 2016 to identify microcephaly cases and conducted a nested case-control study to determine risk factors for microcephaly. Gestational age at birth was estimated based on antenatal ul… Show more

“…3296). Our approach was to screen all stored CSF for CHIKV and link these data to demographic surveillance (KHDSS) for incidence estimation as part of a larger program of work aimed at estimating the case burden of arboviral illnesses in coastal Kenya [ 14 , 24 ]. This study is reported as per the Strengthening the Reporting of Observational Studies in Epidemiology (STROBE) guideline ( S1 STROBE Checklist ).…”

“…Plates were washed with water and left to dry before being scored by eye. Focus reduction neutralizing test (FRNT): This was carried out as previously described (Barsosio et al, 2019) with some alterations. Briefly, serum was diluted in 2-fold steps in triplicate from a starting dilution of 1:20.…”

“…Careful aseptic techniques and training of clinical staff are imperative to optimize sample collection and may improve the validity of results. Aseptic techniques were used during cord blood sample collection to minimise sample contamination, since identification of pathogens associated with adverse maternal and perinatal outcomes was planned 36 , including a recently published study on the association of flavivirus exposure with congenital microcephaly 55 . In addition, cord blood analysis using PCR has mostly focused on vertically transmitted viruses [56][57][58] , and more research on cord blood testing using molecular diagnostics is needed to better understand the clinical significance of detected organisms.…”
