Sperm storageMultiple paternity Microsatellite Conservation A B S T R A C TThe freshwater pond turtle, Emys orbicularis, has recently suffered from population declines throughout its range, mainly due to habitat destruction. The mating strategies of this species were studied using genetic data from successive clutches within and between years. To test for the occurrence and frequency of multiple paternity and sperm storage, genetic paternity at six microsatellite markers was assessed in 114 embryos and hatchlings from single and subsequent clutches of 11 females (including clutches from the same or consecutive years). Multiple paternity was rare and only found in two out of 20 clutches from 11 females. All annual successive clutches and 58% of the clutches in the next year, were fertilized with sperm from the same male. The use of stored sperm is thus a frequent strategy in E. orbicularis. However, hatching rate, hatchling mass, and hatchling length decreased in clutches fertilized by stored sperm, suggesting sperm depletion or deterioration through time. The occurrence of stored sperm despite an associated reduced reproductive output indicated that mating and/or the fertilization process is costly to females. The low incidence of multiple paternity may simply be the residual consequence of the capacity to store viable sperm. These results provide important and innovative insights for the conservation of E. orbicularis. In threatened populations, management strategies may aim to enhance effective copulations in order to increase the reproductive output of females.

Genetic diversity and population genetic structure of the Wood Turtle (Glyptemys insculpta) at Delaware Water Gap National Recreation Area, USA

Glyptemys insculpta is considered to be one of the most endangered freshwater turtles in North America. Here microsatellite markers were employed to investigate the genetic variation and population structure of G. insculpta at Delaware Water Gap National Recreation Area (USA). Seven microsatellites revealed high allelic variation with 13-30 alleles per locus. Observed and expected heterozygosities per locus ranged from 0.875-0.925 to 0.888-0.952, respectively. Pairwise estimates of population structure (h) ranged from 0.000-0.013 to h estimated over all loci and aggregations was not significantly different from zero. Gene flow (Nm) was high and ranged from 19 migrants per generation to infinity in pairwise comparisons. No significant relationship between geographic distance and genetic distance was detected. These data indicate that G. insculpta at DEWA represent a single, genetically diverse management unit for conservation.

Inheritance of nesting behaviour across natural environmental variation in a turtle with temperature-dependent sex determination

Nesting behaviour is critical for reproductive success in oviparous organisms with no parental care. In organisms where sex is determined by incubation temperature, nesting behaviour may be a prime target of selection in response to unbalanced sex ratios. To produce an evolutionary change in response to sex-ratio selection, components of nesting behaviour must be heritable. We estimated the field heritability of two key components of nesting behaviour in a population of painted turtles (Chrysemys picta) with temperature-dependent sex determination by applying the 'animal model' to a pedigree reconstructed from genotype data. We obtained estimates of low to non-detectable heritability using repeated records across all environments. We then determined environment-specific heritability by grouping records with similar temperatures for the winter preceding the nesting season, a variable known to be highly associated with our two traits of interest, nest vegetation cover and Julian date of nesting. The heritability estimates of nest vegetation cover and Julian date of nesting were qualitatively highest and significant, or nearly so, after hot winters. Additive genetic variance for these traits was not detectable after cold winters. Our analysis suggests that the potential for evolutionary change of nesting behaviour may be dependent on the thermal conditions of the preceding winter, a season that is predicted to be especially subject to climate change.
